【摘要】 目的探讨MCP—1启动子区A-2518G基因多态性与血清中MCP-1水平及哈尔滨地区汉族人脑梗死的关系。方法应用聚合酶链反应-限制性片段长度多态性(PCR—RFLP)方法,检测113例脑梗死患者、98例健康对照者MCP-1启动子区A-2518G基因的多态性。应用ELISA方法,检测39例脑梗死患者、38例健康对照者血清中 MCP-1水平。结果各基因型频率和等位基因频率在正常人群及脑梗死患者中无显著性差异(P>0．05)。GG型 GA型的人群血清中MCP-1表达水平较AA型者高(P<0．05)。结论 MCP-1启动子区A-2518G基因多态性可以影响血清MCP-1水平,但它与哈尔滨地区汉族人脑梗死不相关。更多还原

【Abstract】 Objective To investigate the MCP-1 promoter-2518A/G gene polymorphism with serum MCP-1 level and its association with cerebral infarction (CD. Methods The A/G transition polymorphism at position promoter -2518 in the MCP-1 gene was determined by restricted fragment length polymorphism using polymerase chain reaction (PCR-RFLP) method in 113 patients with CI as well as 98 controls. The genotype distribution and allele frequency among different groups were compared. The serum level of MCP-1 was determined by ELISA in 39 patients with CI as well as 38 controls. Results The serum level of MCP-1 from individuals who carry the -2518G allele (G/G + G/A, 396. 02±371. 02pg/ml)was higher than A/A homozygote(A/A,201. 90± 228. 71pg/ml) (P< 0. 05). The serum level of MCP-1 in patients with CI (399. 32±320. 66) did not differ from that of controls(296. 87 ±256. 99)(P>0. 05). The genotype G/G,G/A,A/A distribution of MCP-1 were 33. 6%,53. 1% and 13. 3% in patients with CI,and were 42. 9% ,43. 9% and 13. 3% in controls (P>0. 05). The allele frequency of MCP-1 in patients with CI were 2518G 60. 2%,2518A 39. 8% and 2518G 64. 8%,2518A 35. 2% in controls (P>0. 05). Conclusion The A/G transition polymorphism at position promoter -2518 in the MCP-1 gene affects the serum level of MCP-1. The polymorphism of MCP-1 promoter-2518A/G gene is not significantly associated with cerebral infarction among Han nationality in Harbin China.更多还原
