何首乌制剂在Aβ致海马神经元凋亡中的Bax表达
Effect of polygonum multiflorum thumb on apoptosis of hippocampal neurons induced by Aβ
【摘要】 目的探讨何首乌制剂(PMT)对β-淀粉样蛋白1-40(Aβ1-40)引起的大鼠海马神经元凋亡的影响。方法用Aβ1-40微量注射至大鼠右侧海马,建立AD模型,用何首乌制剂进行干预,于不同时间点分别进行电Y迷宫测试,用免疫组化染色法测定Bax阳性神经细胞数的表达。结果模型组鼠的学习记忆获得能力下降,海马Bax表达增加;何首乌组的学习记忆获得能力有所改善,海马Bax表达下调;但与对照组比仍有差异(P<0.01)。结论 Aβ1-40 注入海马引起大鼠学习记忆损害,可能与其促进Bax的表达有关,何首乌对模型鼠的学习记忆障碍具有保护作用, 可能与其抑制Bax的表达有关。
【Abstract】 Objective To study the effect of Polygonum Multiflorum Thumb (PMT) on apoptosis of hip-pocampal neurons induced by Aβ1-40. Methods The AD (Alzheimer disease) model rats are established by injection of amyloid-beta protein into right hippocampus of rats and are fed with PMT. Learning and memory ability was studied by Y-electric maze at different time. The expression of apoptosis related protein Bax in rats’hippocampus were studied by immunohistochemistry method. Results The experiment showed that the learning and memory a-bilities were worse,the expression of Bax was increased in AD model rats. The expression of Bax was decreased in the PMT’s rats. Learning and memory abilities were better than those of the AD model rats,but were also worse than the control group (P<0. 01). Conclusion The injection of Aβ1-40 into hippocampus of rats can cause the loss of learning and memory abilities, which is related the up-expression of Bax. PMB is found to improve learning and memory abilities and may be related to its inhibition of expression of Bax.
