大鼠急性心肌损伤与CT-1mRNA相关性的实验观察

【摘要】 目的研究心肌营养素1(cardiotrophin-1,CT-1)在异丙基肾上腺素(isoprenaline,Iso)致大鼠急性心肌损伤中的保护作用。方法将Wistar雄性大白鼠随机分为3组:对照组、Iso组和Iso+PD98059组,光镜观察心肌组织的病理变化,生化方法检测心肌酶,用RT-PCR方法检测心肌组织CT-1mRNA的表达。结果光镜下Iso+PD98059组心肌病理损伤程度较Iso组重,且CT-1mRNA表达水平及心肌酶活性明显升高(P<0.05)。结论CT-1具有心肌保护作用,可能与减轻心肌细胞坏死的程度有关。

【Abstract】 Objective To study the protective effects of cardiotrophin-1(CT-1) on rat myocardial injury induced by isoprenaline(Iso). Methods Wistar rats were divided into three groups at random, being control group, Iso group and Iso+ PD98059 group respectively. The pathological changes of myocardium were observed under microscope. Levels of myocardial enzyme were detected by biochemical method. The expressions of CT-1 mRNA were tested with RT-PCR method. Results In the Iso + PD98059 group, myocardial necrosis was more serious than that in the Iso group,. The expression of CT-1 mRNA and the levels of myocardial enzyme were increased obviously(P < 0.01 or P < 0.05)as well. Conclusion CT-1 plays a protective role for myocardium, possibly through alleviating the level of myocardial cell necrosis.
