景天三七中没食子酸和总酚酸含量测定

【摘要】 目的:测定景天三七中没食子酸和总酚酸的含量。方法:没食子酸含量分析采用高效液相色谱法。色谱条件为:流动相为甲醇-磷酸水溶液(pH3.0)(90∶10),检测波长271 nm,流速1.0 mL/m in。总酚酸的含量测定采用可见分光光度法,以没食子酸为对照品,0.6%FeC l3—0.9%K3[Fe(CN)6](1∶0.9)混合溶液为显色剂,在720 nm波长处进行测定。结果:没食子酸分析的线形范围为0.343 0μg~1.200μg(r=0.999 7),平均回收率为97.91%,RSD为1.8%(n=6)。总酚酸分析的线形范围为0.4640~2.320μg/mL(r=0.999 3),平均回收率为99.21%,RSD为2.0%(n=6)。结论:本方法简单快捷,重现性好,可作为景天三七中没食子酸和总酚酸的质控方法。

【Abstract】 AIM: To develop the methods for the quantitative analysis of gallic acid and total phenolic acid in Sedum Aizoon L. METHODS: Gallic acid was analyzed by HPLC on a Hypersil BDS C18 column and detected at 271 nm.The mobile phase was methol-water(adjusted to pH=3.0 with H3PO4)(90∶10)at a flow rate of 1.0 mL/min.Total phenolic acid was analyzed by spectrophotometry at 720 nm.The colour-developing agent was the mixture solution of 0.6%FeCl3—0.9%K3[Fe(CN)6](1∶0.9). RESULTS: Calibration graphs were constructed in the range of 0.343 0-1.200 μg(r=0.999 7) for gallic acid and 0.4640-2.320 μg/mL(r=(0.999 3)) for total phenolic acid.The average recoveries were 97.91%(n=6) for gallic acid and 99.21%(n=6) for total phenolic acid.The RSD of the methods were 1.8% and 2.0%,respectively. CONCLUSION: The methods were fast,reliable,accurate and suitable for analysis of gallic acid and total phenolic acid and quality control in Sedum Aizoon L.
