妊娠相关糖蛋白对E型选择素介导的内皮细胞黏附作用研究
Effect of pregnancy-associated glycoproteins on E-selectin-mediated endothelial cell adhesion
【摘要】 目的:本实验旨在探讨高度纯化的羊水和孕妇血清人绒毛膜促性腺激素(human chrionic gonadotropin,hCG)和glycodelin对E型选择素介导的HepG2细胞与内皮细胞黏附过程的影响。方法:以色谱仪分离和纯化羊水和血清的hCG和glycodelin,进一步测定[51Cr]标记的肝癌细胞系-HepG2细胞在不同来源、不同浓度hCG和glycodelin作用下和经过白细胞介素-1β(IL-1β)刺激后表达E型选择素的内皮细胞的黏附能力,从而评价这两种糖蛋白对E型选择素介导的内皮细胞黏附过程的影响。结果:羊水和血清hCGs能有效抑制E型选择素介导的内皮细胞黏附,其抑制作用呈浓度依赖关系,IC50分别为1·5×10-7mol/L和2·1×10-7mol/L,相对抑制力分别为纯四糖结构sialyl-Lewisx(sLex)的1·5×104和1·1×104倍;羊水和血清glycodelin同样有此作用,其IC50分别为3·2×10-6mol/L和6·3×10-6mol/L,相对抑制力分别为sLex的7·2×102和3·7×102倍;纯四糖结构sialyl-Lewisx(sLex)的IC50为2·3×10-3mol/L。结论:妊娠相关糖蛋白hCG和glycodelin是E型选择素介导的内皮细胞黏附的有效抑制物。
【Abstract】 AIM: To explore the effect of high purified human chorionic gonadotropin (hCGs) and glycodelins isolated from amniotic fluid and serum of pregnant women on E-selectin mediated endothelial cell adhesion. METHODS: The hCGs and glycodelins in human amniotic fluid and serum collected from pregnant women were purified by using chromatography. The binding of ~ 51 Cr-labelled HepG2 cells to IL-1β stimutated human umbilical vein endothelial cells (HUVECs) in the presence of increasing concentrations of hCGs and glycodelins was quantified. The effect of hCG and glycodelin on E-selectin-mediated endothelial cell adhesion was observed. RESULTS: HCGs isolated from amniotic fluid and serum of pregnant women inhibited the E-selectin mediated adhesion of HepG2 cells to HUVECs in a dose-dependent manner, the IC_ 50 was 3.2×10~ -6 mol/L and 6.3×10~ -6 mol/L, respectively and the relative inhibition was 1.5×10~4 and 1.1×10~4, respectively, compared to sialyl-Lewis~x. The IC_ 50 of amniotic fluid glycodelin and serum glycodelin was 3.2×10~ -6 mol/L and 6.3×10~ -6 mol/L respectively, and the relative inhibition was 7.2×10~2 and 3.7×10~2, respectively, compared with sialyl-Lewis~x. CONCLUSION: Pregnancy-associated glycoproteins, hCG and glycodelin, are the powerful inhibitors for E-selectin-mediated endothelial cell adhesion.
