TuMV-Nib反义基因对大白菜的遗传转化研究
Genetic Transformation of Anti-TuMV-Nib into Transgenic Chinese Cabbages
【摘要】 以‘福山包头’大白菜子叶为试材,以Anti-TuMV-Nib为目的基因,利用根癌农杆菌介导法,建立了大白菜再生转化体系,获得转化植株。经PCR-Southern杂交、RT-PCR及ELISA检测,证明外源TuMV-Nib基因已整合到大白菜核基因组中并获得了表达。
【Abstract】 Agrobacterium tumiefaciens-mediated regeneration system was established using Anti-TuMV-Nib gene. Cotyledons with petiole from aseptic seedlings of Chinese cabbage (Brassica campestris ssp. pekinensis‘Fushanbaotou’) were used as explants and the transgenic plants were obtained. The analysis of PCR-Southern blotting, RT-PCR and ELISA detection showed that the target gene was integrated into the Chinese cabbage genome and expressed.
