大鼠脊髓损伤后神经细胞损伤与Caspase-3关系的研究
The relationship of neuronal injury and expression of Caspase-3 after spinal cord injury
【摘要】 目的探讨大鼠脊髓损伤与凋亡促进因子Caspase-3之间的关系。方法SD大鼠24只,脊髓损伤后取损伤脊髓标本,应用HE染色、免疫组织化学和Caspase-3mRNA原位杂交,TUNEL染色,观察脊髓损伤区神经元损伤变化与Caspase-3表达之间的关系。结果脊髓损伤后5h可监测到Caspase-3免疫组化阳性细胞(10.2个/高倍视野),22~48h时达高峰(21.5~24.2个/高倍视野)。Caspase-3mRNA原位杂交阳性表达始于5h(6.66个/高倍视野),24h达高峰(14.55~18.22个/高倍视野),二者均在72h后呈明显下降趋势。损伤后24h,可监测到TUNEL阳性细胞,持续至72h。Caspase-3mRNA与Caspase-3呈正相关,相关系数为0.716(P<0.05).4~16h受损神经原形态基本正常,24 ̄48h细胞凋亡特征明显,72h后,几乎所有神经元形态呈现严重变化。结论大鼠脊髓损伤后神经元发生一系列形态变化,其演变规律与凋亡促进因子Caspase-3有密切相关性。
【Abstract】 Objective To study the relationship between injured neuron and expression of Caspase-3 after spinal cord injury. Methods The injured spinal cord was performed with Histopathologic and HE stain and Caspase-3 protein and mRNA expression level was assessed by immunohistochemistry and in situ hybridization respectively. Results Caspase-3 protein immunohistochemical staining positive cells (10.2/HP) were identitified from spinal cord injured area after 5h and reached the peak at 22~48h(21.5~24.2/HP).In situ hybridizational positive expression cells(6.66/HP) were identified from spinal cord injury after 5h ,which reached the peak at 24 h(14.55~18.22/HP). Caspase-3 mRNA and protein expression level was decreased after 72h.There was a statistically significant correlation between Caspase-3 Mrna and protein expression(r=0.716,P<0.05).TUNEL-positive cells appeared after spinal cord injury from 24 to 72h.The injuried neurons remained normal morphlogic in 4~16h,and the features of cellular apoptosis showed apparently in 24~48h.after 72h almost all of neurons presented severe histopathologic changes. Conclusions our data demonstrated a series of morphologic alteration in spinal cord neurons after acute spinal cord injury, which is consistent with the expression of Caspase-3 mRNA and protein.
