低氧诱导因子-1研究进展
Progress of Hypoxia-inducing Factor 1
【摘要】 低氧诱导因子-1(HIF-1)是机体的一种重要转录因子,其表达和活性受到细胞氧浓度的严密调控。它通过控制血管生成来调节氧的运输能力,通过调控糖酵解作用来增强机体对低氧的耐受。肿瘤的生长和血管生成与HIF-1的表达紧密相关,抑制HIF-1活性可能是治疗癌症的一种途径。另外,对HIF-1表达水平的药理学控制,可能也是治疗慢性肺疾患、脑缺血和心肌缺血等疾病的一种新思路。
【Abstract】 Hypoxia-inducing factor 1(HIF-1) is an essential transcription factor,whose expression and activity are tightly regulated by cellular oxygen concentration.HIF-1 controls oxygen delivery via angiogenesis and promotes the tolerance to hypoxia via regulating glycolysis.Tumor growth and angiogenesis are correlated with the expression of HIF-1,therefore,inhibiting the activity of HIF-1 may be a strategy for cancer therapy.Besides,Pharmacologic manipulation of HIF-1 in its expression level may provide a novel therapeutic approach to chronic lung disease,cerebral and myocardial ischemia,and so on.
