组织微阵列技术在地塞米松诱导SAP大鼠胰腺细胞凋亡中的应用
Application of Tissue Microarrays in Apoptosis of Acinar Cell in pancreas of Rat with Severe Acute Pancreatitis Induced by Dexamethasone
【摘要】 目的应用组织微阵列技术研究地塞米松对SAP细胞凋亡及其调控基因的影响。方法用改良Aho法制备SAP大鼠模型,模型组、地塞米松治疗组、假手术组各45只。术后3、6、12h观察各组存活率、胰腺病理变化、Bax、Bcl-2表达,凋亡细胞种类及凋亡指数。结果模型组、治疗组存活率无差异(P<0·05);治疗组胰腺病理改变较模型组轻;治疗组Bax各时点阳性率高于模型组(P<0·01);模型组12h胰头部Bcl-2高于治疗组(P<0·01);治疗组6h凋亡指数高于模型组(P<0·01)。结论地塞米松治疗SAP可能与凋亡调控基因Bax、Bcl-2诱导的胰腺细胞凋亡有关;组织微阵列技术在胰腺炎病理检测中优势明显。
【Abstract】 Objective To apply tissue microarrays to study the influence of dexamethasone on apoptosis of SAP and expression of Bax and Bcl-2. Methods Rats SAP models were Prepared via improved Aho, There are 45 rats each in model group, treatment group,false operation group. The survivals, pathological changes, Bax and Bcl-2 expression, apoptotic cell category and apoptotic index changes were observed at 3h, 6h and 12h after operation in each group. Results There was no apparent difference between model group and dexamethasone group in mortality(P>0.05); The pathological changes of the treatment group was less than that in model group; The positive rates of Bax in treatment group at all time points were higher than that in model group (P<0.01); The positive rate of Bcl-2 in pancreas head in model group was higher than that in treatment group at 12h(P<0.01); The apoptotic index of the treatment group was higher than that in model group in 6h(P<0.01). Conclusions The mechanism of dexamethasone in treating SAP is possibly related to the apoptosis induced by apoptotic regulating genes like Bax and Bcl-2; The advantages of tissue microarrays in pancreatitis pathological examination is obvious.
