邻苯二甲酸二异辛酯致小鼠肝DNA蛋白质交联效应研究
Study on DNA-Protein Crosslinks of Mice Liver Induced by Di-(2-ethylhexyl)Phthalate
【摘要】 邻苯二甲酸酯是一类广泛应用于人类生产和生活的酸酞酯类人造化学物,许多研究表明其具有广泛的生物学毒性。DNA-蛋白质交联(DNA-proteincrosslinks,DPC)是DNA分子的一种严重损伤。本文以小鼠肝为实验材料,采用Kcl-SDS沉淀法检测邻苯二甲酸二异辛酯(DEHP)腹腔注射染毒两周后,检测其导致的小鼠肝DPC效应。实验结果表明,低浓度的DEHP(62.5mg/kg)不能致DPC效应(P>0.05),随着染毒浓度的倍增(125mg/kg,250mg/kg,375mg/kg),其DPC系数也随之升高,可导致明显的DPC效应(P<0.01),而在500mg/kg染毒组,由于其染毒剂量达到小鼠的致死水平,其DPC效应虽然与空白对照组相比有极显著性差异(P<0.01),但<125mg/kg染毒组。结论DEHP在低浓度(62.5mg/kg)时不引起小鼠肝的DPC效应,但其DPC系数随着染毒浓度(125mg/kg,250mg/kg,375mg/kg)的升高而升高,可以导致明显的DPC效应,在染毒剂量(500mg/kg)达到致死水平的染毒组DPC系数明显降低,但仍明显高于空白对照组。
【Abstract】 Phthalates are a family of manufactured chemicals that are widely used in our daily life. Many researches have indicated that it has extensive toxicities. DNA-protein crosslinks (DPC) is a kind of serious DNA damage。To study the effect of di-ethylhexyl phthalate (DEHP) induced DPC in mice liver. This study took mice liver as experimental material and made use of KCl-SDS assay to deuermine the amount of DPC induced by DEHP with celiac injection in vivo for two weeks. From the KCl-SDS assay results DEHP can not cause DPC at low levels (62.5mg/kg, P>0.05), but DPC coefficient increases along with concentration of DEHP (125 mg/kg, 250 mg/kg, 375 mg/kg) rising, and it can cause DPC significantly (P<0.01). 500mg/kg group can cause DPC significantly (P<0.01) compared with the control group, but its DPC coefficient is lower than 125 mg/kg group because concentration of DEHP is at level of lethal dosage in mice. From the above results we can conclude that DEHP can not cause DPC at low levels (62.5mg/kg), but it can cause DPC significantly at mid-high levels (125 mg/kg, 250 mg/kg, 375 mg/kg). DPC coefficient of the group at lethal dosage decreases significantly, but it is still higher than that in control group.
