【摘要】 目的观察低氧对结肠癌SW480细胞中NDRG1、MMP-2蛋白表达的影响。方法分别在常规(常氧组)和低氧(低氧组)条件下培养结肠腺癌SW480细胞。运用免疫细胞化学染色法检测2组NDRG1蛋白表达,并用图像分析仪测光密度值(OD值);ELISA法检测MMP-2表达。结果低氧组细胞NDRG1呈强阳性表达,而常氧组表达较弱,低氧组细胞NDRG1蛋白表达水平(0．1634-4-0．0079)显著高于常氧组(0．1358±0．0027)(P<0．05);MMP-2表达也显著高于常氧组(P<0．01)。结论低氧诱导了SW480细胞NDRG1、MMP-2表达上调。更多还原

【Abstract】 Objective To observe effects of hypoxia on the expression of NDRG1 and MMP-2 in colon cancer cell SW480.Methods NDRG1 protein was measured by immunocytochemistry, and MMP-2 protein was measured by ELISA. Results The level of NDRG1 proteins expression in SW480 cells was significantly higher in the hypoxia group than in the normoxia group (P<0.05).The results of ELISA revealed that the level of MMP-2 protein expression in culture medium was significantly higher in the hypoxia group than in the normoxia group (P<0.05).
