KAI1基因抑制人胰腺癌细胞转移的体内实验
KAI1 gene inhibits in vivo metastasis of human pancreatic cancer.
【摘要】 目的研究体内直接注射KAI1基因抗胰腺癌转移作用。方法MiaPaCa胰腺癌细胞裸鼠皮下接种后,随机分为2个大组(接种后第10天和第21天治疗组),每组再分别瘤内注射pCMV-KAI1重组质粒100μg,pCMV-KAI1-脂质体(50μg∶50μg)和生理盐水,每7d注射1次,共注射3次。于第50天观察KAI1对胰腺癌生长转移影响。结果接种后10d进行基因注射组中,KAI1治疗组和脂质复合物治疗组肿瘤体积、瘤重、肺重、肝重和肺表面转移结节数均小于对照组(P<0.05),体积抑瘤率分别为63.79%和73.51%,瘤重抑瘤率分别为77.12%和83.26%,肺重分别为(0.33±0.09)g和(0.30±0.09)g,肝重分别为(1.01±0.27)g和(0.99±0.21)g,对照组肺肝重分别为(0.45±0.09)g和(1.62±0.39)g,对照组肺表面转移结节数平均为(2.33±1.63)个,治疗组结节数仅为(0.5±0.83)个。pCMV-KAI1治疗组与脂质复合物治疗组相比差异无显著性(P>0.05);接种后21d进行基因注射组中,治疗组肿瘤体积、瘤重、肺重、肺转移灶和肝重与对照组相比差异无显著性(P>0.05)。结论KAI1基因瘤内注射具有抑制MiaPaCa胰腺癌生长及转移作用,KAI1基因对已成瘤的胰腺癌生长的抑制作用与瘤内给药时间密切相关。
【Abstract】 Objective To evaluate the role of KAI1 gene injection in inhibiting metastasis of pancreat ic cancer in vivo . Methods Heterotopic human pancreatic ade noca rcinoma was successfully established in the groin of BALB/C nude mice by subcuta neous injection of MiaPaCaⅡ pancreatic cancer cells. The tumor-bearing mice we re randomly divided into two groups: day-10 group, where treatment was started from the 10th day of cancer cell inoculation, and day-21 group, where tre atment was started from the 10th day of cancer cell inoculation. 100 μg r ecombinantplasmid pCMV-KAI1, 100 μg pCMV-KAI1-LipofectAMINE(1∶1), or 100 μl saline was directly injected into pancreatic adenocarcinoma at a 7-day inte rval for three times. At day 50, the tumor growth and metastasis were measured. Results In the KAI1 group and KAIl-LipofectAMINE group of day -10 group, the tumor volume inhibitory rate of treatment group and control grou p was 63.79% vs 73.51%, the tumor weight inhibitory rate was 77.12% vs 8 3.26%, the weight of lung was (0.33 ± 0.09)g vs (0.30 ± 0.09)g, the weight o f liver was (1.01 ± 0.27)g vs (0.99 ± 0.21)g, and the lung metastatic nodule num ber was 0.5 ± 0.83 vs 2.33 ± 1.63, respectively. These data were less than those of control group( P < 0.05). There was no significant difference between KAI1 group and KAIl-LipofectAMINE g roup ( P > 0.05). In 21-day group, ther e was no significant difference in tumor volume, tumor weight, weight of lung, weight of liver and the lung metastatic nodule number between gene therapy group and salin e control group ( P > 0.05). Con clusions KAI1 gene directly injected into pancreatic adenocarcinoma pla ys an important role in inhibiting the growth and metastasis of MiaPaCaⅡ pancre atic cancer. The suppressive role of KAI1 gene was closely correlated with the t ime phase of injecting gene into tumor.
