Graves病中调节性T细胞功能的改变

【摘要】 目的探讨Graves病发生的免疫机制,了解调节性T细胞在其发病中的作用。方法流式细胞术检测32例Graves病(GD组)和20例正常人(正常对照组)中CD4+CD25+T细胞的比例。免疫磁珠法(MACS)分离CD4+CD25+细胞,酶联免疫吸附(ELISA)法检测IL-10的水平,逆转录-聚合酶链反应(RT-PCR)法检测FOXP3的表达。结果与正常对照组相比,GD组CD4+CD25+T细胞数量无明显改变(P=0.804),而FOXP3表达减少(24 h和48 h P均<0.01),IL-10分泌减少(24 h和48 hP均<0.01)。结论GD患者调节性T细胞在外周血淋巴细胞中的比例虽无明显改变,但其功能明显减弱,可能是导致其发病的原因之一。

【Abstract】 Objective To search the immune-mechanism of Graves disease,and get known the function of regulatory T cell in its occurrence.Methods Flow cytometry were used to detect the proportion of CD4+CD25+ T cells in 32 Graves disease patients(GD group) and 20 healthy volunteers(control group).MACS were used to isolate CD4+ CD25+ T cells.RT-PCR was used to detect the expression of FOXP3 of them,and ELISA to detect the level of IL-10.Results There was no significant change of proportion of CD4+CD25+ T cells between GD group and control group(P=0.804),while expression level of FOXP3 mRNA and secretion level of IL-10 in GD group were lower than those in control group(P<0.01 or P<0.05).Conclusion Function of regulatory T cells was significantly weakened though its amounts were not changed.This may play an important role in the occurrence of Graves disease.
