强力霉素对人角膜上皮细胞FasL表达的影响
Effect of doxycycline on expression of FasL in human corneal epithelial cells
【摘要】 目的观察正常人角膜FasL蛋白的表达情况,评价人角膜上皮细胞FasL蛋白对强力霉素反应的表达特性。方法使用免疫组织化学染色观察正常人角膜上皮FasL的表达;用组织块培养法培养人角膜上皮细胞,将得到的细胞随机分为两组,一组使用含200μg/ml强力霉素的培养液,另一组不含强力霉素,分别再培养24h。运用流式细胞技术测定24h后FasL的表达情况。结果FasL在正常人角膜上皮细胞中呈阳性表达;经强力霉素干预的角膜上皮细胞FasL的表达高于未经强力霉素干预的角膜上皮细胞(P<0·01)。结论正常人角膜上皮细胞有FasL的表达;强力霉素可使体外培养的人角膜上皮细胞FasL的表达增高,有望降低临床角膜上皮细胞移植后的免疫排斥反应。
【Abstract】 ObjectiveIn recent years,people found that the immune privilege have a close relation to Fas/FasL.The expression of Fas ligand(FasL) in corneal epithelium and endothelium plays an important role in elevating the successul rate of corneal transplantation.This experiment detected the expression of FasL in normal human corneal epithelial cell and evaluated the influence of doxycyline on expression of FasL.MethodsHuman corneal epithelial cells were cultured in SHEM culture media.The cultured cells were randomly assigned to 200μg/ml doxycycline-treated group and untreated group.Then flow cytometry was used to analyze the fluorescence index of FasL after 24hours.Immunohistochemistry was used to detect FasL protein in normal human corneal epithelial cell.ResultsNormal human corneal epithelial cells were stained into brown in cell membrane and endochylema,showing the positive expression of FasL.In doxycycline-treated group,the fluorescence index(FI)of FasL was significantly enhanced compared with untreated group(1.239±0.285 versus 0.840±0.100),indicating the increase of FasL expression on corneal epithelial cell.(P= 0.007 ).ConclusionFasL is expressed in normal human corneal epithelial cell.Doxycycline induces the increase of FasL expression in vitro,suggesting a potential effect of prolonging allograft survival and resulting in a modest increase in allograft acceptance.
