聚四氢呋喃聚乳酸共聚物C4对眼组织毒性的实验研究
An experimental study of poly(tetrahydrofurfuryl-co-lactic acid) copolymer toxicity on ocular tissue
【摘要】 目的研究聚四氢呋喃聚乳酸共聚物(C4)对眼组织的毒性。方法将聚四氢呋喃聚乳酸共聚物分别置入兔眼玻璃体腔和前房内,定期进行临床观察和视网膜电图检查,6个月后摘除眼球行病理观察。结果聚四氢呋喃聚乳酸共聚物在兔眼玻璃体腔内(168.3±12.6)d完全降解,在前房内(136.5±9.6)d完全降解,视网膜电图正常,眼组织病理观察无异常,兔眼压于手术后第3d及第120d左右两次出现升高。聚合物直接接触角膜内皮1.5个月后内皮细胞消失。结论聚四氢呋喃聚乳酸共聚物对视网膜、晶状体无明显毒性,但不能直接接触角膜内皮。
【Abstract】 ObjectiveTo study the ocular tissue toxicity after poly (tetrahy drofurfuryl-co-lactic acid) Copolymer (PTHF-PLA) was implanted into rabbit eye.Methods0.4ml of PTHF-PLA (C_ 4) was placed into vitreous cavity in 45 rabbits in group A,and 0.05ml of C_ 4 was used for the placement of anterior chamber in 45 rabbit in group B,and 10 rabbits was used as control.The ocular pressure,electroretinogram(ERG) and other clinical examination were performed on all the rabbits.The eyes were then enucleated and processed for histological evaluation after six months.ResultsThe degradation time of C_ 4 was ( 168.3±12.6) days in the vitreous cavity and that in the anterior chamber was(136.5±9.6)days and the concentration was negligible after 136 days.The b wave of ERG was 214.46±17.31,223.53±26.45 and 215.73±15.36μV at 1 month,2 and 6 months in the A group without statistically significant difference in comparison with those of control group(215.39±22.56,227.92±19.62 and 211.87±23.18,respectively ).No marked ocular tissue abnormalities were observed under the light or electromicroscope.Intraocular pressure was significantly higher at day 3 (16.24±1.36) and day 120 (16.53±1.16)after operation in comparison with that before operation (11.42±1.36) and other time point (13.69±1.52)of postoperation in the group A.The corneal endothelia cells were disappeared 1.5 months after copolymer contacted with cornea.ConclusionNo retinal and lens toxicity was seen in C_ 4 filled eyes in rabbits.The direct contact of C_ 4 with cornea of rabbits can damage corneal endothelia in the experimental model.
