蛋白酶体抑制剂PS-341对多发性骨髓瘤间充质干细胞IL-1β表达的影响
Effects of proteasome inhibitor PS-341 on the expressions of cytokine IL-1β of mesenchymal stem cells in patients with multiple myeloma
【摘要】 目的初步探讨蛋白酶体抑制剂PS341在治疗多发性骨髓瘤(MM)时对间充质干细胞(MSC)细胞因子白介素(IL1β)分泌的影响。方法10%胎牛血清(FBS)的1640培养液培养MSC,细胞数达0.5~1.0×106时以终浓度50nmol/L的PS341作用4h,RTPCR检测细胞因子IL1β表达。结果PS341作用后MSC分泌的IL1β表达有显著下降,完全缓解及经PS341治疗后患者的IL1β未见表达;与难治复发组比较,PS341对完全缓解患者MSCIL1β的表达抑制更明显。结论PS341抑制MM患者MSC细胞因子IL1β的表达。
【Abstract】 Objective To explore the effect of proteasome inhibitor PS-341 on the expressions of cytokine IL-1β of mesenchymal stem cells(MSC) in the patients with multiple myeloma(MM). Methods MSC were cultured in medium of 1640 containing 10% FBS and exposured in 50nmol/l PS-341 for 4h when cells developed to (0.5~1)×10~6.The cells were harvested and detected the expression of cytokine of IL-1β by RT-PCR.Results The expression of cytokine IL-1β of MSC decreased remarkablely after treated with PS-341. The IL-1β expression was not measured in the patients who were in complete remission(CR) or treated with PS-341.Compared to refractory/relapsed group,the expression of MSC IL-1β decreased more significantly in CR group. Conclusion Proteasome inhibitor PS-341 inhibits the expressions of cytokine IL-1β of mesenchymal stem cells(MSC) in MM patients.
