银杏叶总黄酮介导的豚鼠腹腔神经节细胞的去极化
Total flavone ginkgo-mediated depolarization in the neurons of guinea pig celiac ganglia in vitro
【摘要】 目的观察并分析银杏叶总黄酮(TFG)对豚鼠离体腹腔神经节(CG)细胞膜电位的影响。方法应用细胞内生物电记录技术和离体神经节灌流给药方法。结果TFG可使45·8%的CG细胞去极化,幅度和时程分别为(7·9±0·7)mV和(245·9±29·2)s,且表现剂量依赖性;10%超极化;另有10%出现先超极化后去极化的双相反应;还有34%几乎不发生反应。多数细胞的去极化可被低钙/高镁Krebs液部分阻断,但不受胆碱和肾上腺素受体阻断剂的影响。在TFG诱发的CG细胞去极化的同时伴膜电阻减小(47·2%)、增大(21·8%)或不变(30·9%);无钠或无钙的Krebs液可部分阻断CG细胞去极化,而四乙基铵则使细胞去极化增强。结论TFG可诱发豚鼠CG大多数细胞的去极化,其机制可能与Na+、Ca2+内流增强和K+外流减弱有关。
【Abstract】 Objective To investigate and analyze the effect of the total flavone ginkgo (TFG) on membrane potential of the neurons in celiac ganglia (CG) of guinea-pig in vitro. Methods Intracellular microelectrode recording and administration of superfusion were used. Results Superfusion of celiac ganglia with TFG evoked a concentration-dependent membrane depolarization in 45.8% of the neurons, amplitude and duration are 7.9 mV±0.7 mV and 245.9 s±29.2 s respectively and 10% demonstrated hyperpolarization and 10% biphasic response which consisted of an initial hyperpolarization phase and a followed depolarization, also 34% no response to TFG. The depolarization of most neurons could be blocked partly by cholinoceptor (atropine and hexamethonium) and adrenoceptor antagonist (phentolamin and propranolol); Among the depolarization neurons, the membrane resistance (Rm) decreased in 47.2% increased in 21.8% and unchanged in 30.9%; The depolarization can be blocked partly by Na~+ and Ca~ 2+ free Krebs solution and depolarization increased by traethylammonium (TEA). Conclusion TFG can evoke the depolarization of most CG neurons, and the mechanism probably is associated with the increasing influx of Na~+ and Ca~ 2+ and the decreasing outflow of K~+.
