丘脑网状核内一氧化氮能神经元对大鼠睡眠-觉醒周期的影响
Effect of nitric oxide neurons in reticular thalamic nucleuson sleep-waking cycle in rats
【摘要】 目的研究丘脑网状核(RT)一氧化氮能神经元对大鼠睡眠-觉醒周期的影响。方法采用脑立体定位、核团埋管及微量注射和多导睡眠描记技术(PSG)。结果RT双侧分别微量注射一氧化氮合酶抑制剂左旋硝基精氨酸2·0μg后引起睡眠增多;注射一氧化氮的前体左旋精氨酸0·5μg后对睡眠有改变,但无显著性意义,其促觉醒作用未能被左旋硝基精氨酸所阻断;注射一氧化氮的供体硝普钠0·2μg后可明显增加觉醒时间,缩短睡眠时间,并且可以阻断左旋硝基精氨酸的促睡眠作用。结论RT参与大鼠睡眠-觉醒周期的调节,RT中的一氧化氮能神经元可能有促觉醒作用。
【Abstract】 Objective To study the effects of nitric oxide(NO) neurons in reticular thalamic nucleus(RT) on sleep-waking cycle in rats.Methods Brain stereotaxic technigne,microinjection,polysomnography(PSG) and nucleus spile.Results N-nitro-L-arginine (L-NNA , 2.0 μg) ,aninhibitor of nitric oxide synthase (NOS), increased the sleep time of slow wave sleep(SWS) and total sleep time(TST); L-arginine (L-Arg, 0.5 μg) , a precursor of NO,had an effect on sleep-wakestate, but no significance. the wake-promoting effect of L-Arg cannot be blocked by L-NNA; Sodium nitrioprusside (SNP, 0.2 μg) , the donor of NO,caused enhancement of wakefulness and can block the sleep-promoting effect of L-NNA.Conclusion RT plays an important role in sleep-waking cycle in rats; nitric oxide neurons in reticular thalamic nucleus can promote wake.
