【摘要】 目的研究Blipam对脑缺血损伤的保护作用。方法采用小鼠断头法、耐缺氧法观察Blipam对缺血性脑损伤的影响;建立大鼠局灶性(MCAO)脑缺血模型,观察Blipam对大鼠行为学、脑梗塞重量的影响,并测定缺血侧脑组织中丙二醛(MDA)、一氧化氮(NO)含量及乳酸脱氢酶(LDH)活性。结果Blipam能显著延长小鼠断头后喘息持续时间和耐缺氧时间;Blipam减轻MCAO大鼠脑组织梗塞重量,抑制MCAO大鼠异常神经症状的产生,同时降低脑组织中MDA含量,提高脑组织中NO水平,升高脑组织中LDH的活性。结论Blipam对缺血缺氧性脑损伤具有显著保护作用,其机理可能与提高脑组织中NO含量、抑制脂质过氧化有关。

【Abstract】 Objective To study the protective effects of Blipam against cerebral ischemia injury. Methods Decapitation method and close hypoxia method were used to observe the effect of Blipam against cerebral ischemia injury. The permanent occlusion of the proximal of the right middle cerebral artery(MCA) was used for making the focal cerebral ischemia models. The extent of neurological deficits was observed, and the infarct area was measured using NBT staining technique. The activity of LDH and the content of MDA and NO in the ischemic cerebral cortex were determined. Results Blipam prolonged the grasping time after decapitation and the survival time after anoxia in mice. Blipam significantly decreased the infarct weight and ameliorated neurologic deficits score of MCA occlusion rats. Blipam decreased MDA content and increased NO content and increased the activity of LDH of the ischemic cerebral cortex.Conclusion Blipam has protective effects against cerebral ischemic injury,while it may be related to increasing NO production and inhibiting free radicals of the ischemic cerebral cortex.
