大鼠骨髓间充质干细胞在体外向神经元样细胞的分化
Differentiation of rat mesenchymal stem cells into neuron-like cells in vitro
【摘要】 目的探讨2-ME与BHA对大鼠骨髓间充质干细胞(MSCs)分化为神经元样细胞的作用。方法大鼠股骨中分离MSCs,贴壁法体外扩增、传代。以二巯基乙醇(2-ME)预诱导,再加入2-ME、3-叔丁基-4羟基茴香醚(BHA)诱导。免疫细胞化学染色检测巢蛋白(nestin)、神经元特异性烯醇化酶(NSE)、微管联合蛋白-2(MAP-2)、胶质纤维酸性蛋白(GFAP)、乙酰胆碱酯酶(AchE)、γ-氨基丁酸(GABA)、酪氨酸羟化酶(TH)的表达,以鉴定分化细胞的表型特征。尿嘧啶脱氧核苷(BrdU)作为细胞分裂标记检测MSCs增殖特性。结果细胞诱导后具有神经细胞的形态,并被BrdU标记,nestin、NSE、MAP-2、AchE、GABA表达阳性,而GFAP、TH表达为阴性。结论2-ME和BHA在体外能够有效的诱导大鼠骨髓间充质干细胞分化为神经元样细胞。
【Abstract】 Objective To investigate effects of 2-ME/BHA on rat mesenchymal stem cells’(MSCs) differentiation into neuron-like cells in vitro. Methods MSCs were separated from femur marrow of rats and planted in plastic culture capsule,then cultured and expanded in vitro. MSCs were treated with 2-ME, then treated with 2-ME/BHA to induce the differentiation into neuronal-like cells. The phenotypic characteristics of the differentiated cells were examined with observing morphology and immunocytochemistry of nestin, NSE,MAP-2,GFAP, AchE, GABA and TH. The multiplication ability of the MSCs was identified by BrdU labelling. Results The MSCs were induced into neuronal-like cells by 2-ME and BHA treatment, and the differentiated cells showed morphological characteristics of neuronal cells and were labelled by BrdU. Nestin, NSE,MAP-2, AchE and GABA ,but not GFAP and TH were expressed in the neuronal-like cells. Conclusion MSCs can be induced to differentiate into neuronal-like cells by 2-ME and BHA.
