复方菝葜颗粒诱导非小细胞肺癌A549细胞凋亡的实验研究
Research of compound rhizoma smilacinus granule induces apoptosis in Non-Small cell lung cancer A549 cell lines
【摘要】 目的通过中药血清药理学方法,观察复方菝葜颗粒(compoundrhizomasmilacinusgranule,CRSG)对非小细胞肺癌(NSCLC)A549凋亡的影响,探讨CRSG抑制非小细胞肺癌的作用机理。方法雄性SD大鼠30只,随机分为两组,每组15只。一组灌服复方菝葜颗粒,2次/d,另一组灌服等剂量的蒸馏水,第6次灌胃后的2h提取血清。A549细胞株随机分为中药实验组、西药对照组、正常对照组,分别加入10%的含药血清、顺铂3μg/m(LDDP)、10%的SD大鼠血清,培养48h后收集细胞,电镜观察细胞形态,流式细胞仪检测细胞凋亡率。结果各组细胞培养48h后中药实验组、西药对照组细胞数与正常对照组比较明显减少,差异均有显著性(P<0.01);电镜下中药实验组、西药对照组均可见到凋亡小体,正常对照组细胞形态正常;中药实验组、西药对照组细胞凋亡率明显高于正常对照组,相比差异有显著性(P<0.01),中药实验组与西药对照组的凋亡率差异无显著性(P>0.05)。结论复方菝葜颗粒具有减慢细胞增殖速度,并诱导A549细胞凋亡的作用,这可能是其对非小细胞肺癌发挥抑瘤作用的机制之一。
【Abstract】 [Objective] To evaluate the effects of Compound Rhizoma Smilacinus granule(CRSG) on apoptosis in A549 cell lines and its mechanisms upon non-small cell lung cancer by the means of serum pharmacology of Chinese medicine.[Methods] The SD rats were randomly differentiated into two groups(n=15/each group),one administrated with CRSG,another same dose of distilled water.The herbage-containing serum was obtained two hours after the sixth treatment.Non-small lung cancer A549 cell lines were randomly divided into CRSG group,DDP group and normal control group which were cultivated with 10% herbage-containing serum,DDP(3μg/mL) and 10% SD rats serum respectively.The cultivated cells were collected after 48 hours,then electron microscope(TEM) was used to to observe the shape of the cells,flow cytometry assay to detect the rate of apoptosis.[Results] The cell count in CRSG group and DDP group was remarkably less than that in normal control group after 48h treatment(P<0.01).The apoptosis body was observed under the TEM in CRSG group and DDP group after 48h,while the cellular shape held normal in normal control group.CRSG group and DDP group had markedly increased in apoptosis rate than normal control group(P <0.01),while CRSG group and DDP group had no significant difference(P >0.05).[Conclusions] CRSG can Slow down the cellular proliferation and induce the cellular apoptosis,which probably is the part of mechanism of CRSG in inhibiting the growth of non-small cell lung cancer.
