应激系统对缺血预适应心肌细胞凋亡的影响

【摘要】 目的探讨应激系统对缺血预适应中心肌细胞凋亡的影响。方法以家兔在体心经典缺血预适应(IPC)实验为基础,增设结扎肾上腺组(Lig组),米非司酮组(皮质醇受体水平阻滞剂)(MF组),甲基强的松龙组(MP组),以TUNEL法,DNAladder及流式细胞仪(FASCan)检测凋亡的存在及凋亡率,同时检测血清皮质醇动态变化,考察应激启动的皮质醇分泌对缺血预适应心肌细胞凋亡的影响。结果TUNEl染色显示MP组和IPC组比较凋亡率差异没有显著性意义(5.05±2.25%vs3.46±0.82%,P>0.05),MF组和Lig组与IPC组比较差异有显著性意义(7.73±3.08%,16.62±12.35%vs3.46±0.82%,P<0.05);DNAladder半定量显示MP组和IPC组电泳条带明显减少;流式细胞仪检测显示Lig组,MF组心肌细胞凋亡较IPC组差异有显著性意义(3.82±1.38%,3.22±0.67%vs1.72±0.29%,P<0.05),MP组与IPC组比较差异没有显著性意义(2.04±0.74%vs1.72±0.29%)。结论从细胞凋亡角度观察IPC的心肌保护作用,结扎肾上腺,受体水平阻滞皮质醇作用可明显削弱其保护作用,MP可一定程度模拟其保护作用,说明应激系统的部分组分通过影响IPC的心肌细胞凋亡起到其保护作用。

【Abstract】 [Objective] To study the act of stress system on cardiomyocyte apoptosis of ischemic preconditioning(IPC).[Methods] Based on classic IPC research operation using rabbit heart,5 groups were set with 3 adding groups.(Bilateral ligated adrenal group(Lig),mifiprestone group(MF),and a methylprednisolone group(MP).using TUNEL dyeing method,DNA ladder and FACScan to study the rate of apoptosis of the ischemic cardiomyocyte.[Result] By TUNEL dyeing,the difference of apoptosis ratios between MP and IPC group was insignificant(5.05±2.25% vs 3.46±0.82%,P >0.05),comparing with IPC group,the difference of MF and Lig group was significant(7.73±3.08%,16.62±12.35% vs 3.46±0.82%,P <0.05).by FACScan,the difference of apoptosis ratio between IPC and MP group was also insignificant(2.04±0.74% vs 1.72±0.29%,P >0.05),comparing with IPC group,the difference of Lig group and MF group was significant(3.82±1.38%,3.22±0.67% vs 1.72±0.29%,P <0.05),and the DNA ladder also showed a decrease in IPC and MP group comparing with IR,Lig,and MF group.[Conclusion] stress system plays an important role during the IPC process on cardiomyocyte apoptosis.
