改良去细胞同种异体神经移植后免疫反应的观察
Observations of T lymphocyte Subpopulations in Rejection after acellular allografting made by freeze-thrawed combined chemical extraction
【摘要】 目的观察冻融联合改良化学法去细胞同种异体神经植入受体后体内细胞免疫变化从而判断其免疫原性。方法30只雄性Wistar大鼠分成冻融法联合改良化学法供体组(实验组)15只、新鲜异体神经供体组(对照组)15只。另取30只Sprague Dawley大鼠随机分成冻融法联合改良化学法受体组15只,新鲜异体神经受体15只。右侧坐骨神经造成1cm长缺损,用供体组的两种不同方法处理的神经分别与相应受体组进行移植物桥接。植入4周后,将移植段取出,作组织学观察、免疫组化染色,记数上述CD4+、CD8+淋巴细胞变化。结果CD4+、CD8+淋巴细胞变化新鲜异体神经组明显多于冻融法联合改良化学法(P<0.01)。结论经冻融联合改良化学法制备的去细胞同种异体神经具有免疫原性低,作为同种神经移植物不会被排斥吸收。
【Abstract】 [Objective] To observe the change of cellular immunity in vivo after the freeze-thrawed combined optimized acellular nerve was implanted into the recipient so as to judge its immunogenicity. [Methods] 30 Wist cat were divided into freeze-thrawed combined optimized acellular nerve group (FTOAN) and fresh nerve group. The sciatic nerves on the right side of all animals were exposed and 10 cm long segments were removed from the mid-thigh level and replaced by one of the three types of grafts of Sprague-Dawley cat. The The acellular nerve dissected for the histological observation, and the cells of CD4+, CD8+ T lymphocytes were counted by immunohistochemical staining 4 weeks after implantation. [Results] Tlymphocytes in local tissues of the fresh nerve group was geater than in the freeze-thrawed combined optimized acellular nerve group (P <0.01). [Conclusion] The freeze-thrawed combined optimized acellular nerve is low immunogenicity after implantation , They will not be rejected or absorbed by the host after repair nerve defect.
