银杏叶提取物预处理对大鼠肝移植TNF-α mRNA和FasmRNA表达的影响

【摘要】 目的探讨银杏叶提取物(GinkgoBilobaextract,EGb)预处理对大鼠肝移植的保护作用及其对供肝TNF-αmRNA、FasmRNA表达的影响。方法雄性SD大鼠分别作供体、受体;采用Kamada’s袖套法建立大鼠原位肝移植模型,根据供肝切取前1h是否静脉注射银杏叶提取物(40mg/kg)将大鼠随机分为银杏叶提取物预处理组(EGb);生理盐水对照组(NS);假手术组(SO)。分别于供肝再灌注后2、6、24h处死动物,检测血清ALT、AST。部分肝组织,10%的甲醛固定、石蜡包埋,病理切片HE染色作组织学检查,新鲜肝组织用RT-PCR检测TNF-αmRNA及FasmRNA的表达。结果供肝再灌注2、6、24h各时点,银杏叶提取物预处理组大鼠血清ALT水平均明显低于生理盐水对照组(P<0.05)。血清AST水平在供肝再灌注2、6h各时点明显现低于生理盐水对照组(P<0.05)。供肝再灌注后2、6h银杏叶提取物预处理组TNF-αmRNA的表达明显低于生理盐水对照组(P<0.05)。供肝再灌注后2、6、24h银杏叶提取物预处理组FasmRNA的表达明显低于生理盐水对照组(P<0.05)。结论银杏叶提取物预处理大鼠供肝,可以减轻移植肝的缺血/再灌注损伤,影响TNF-αmRNA,FasmRNA的表达,对供肝有保护作用。

【Abstract】 [Objective] To investigate the protective effects and the impact on the expression of TNF-αmRNA and FasmRNA by extract of Ginkgo Biloba leaves preconditioning in rat liver transplantation. [Methods] Male Sprague-Dawley rats were used as donors and recipients of orthotopic liver transplantation(OLT) and were divided into EGb group and NS control group randomly according to whether extract of Ginkgo Biloba leaves was injected by venous (40 mg/kg) 1h before liver grafts harvest. The third group was sham operation(SO). The animals were killed at 2, 6, 24 h after graft reperfusion respectively. Plasma samples were collected for ALT and AST tests. Fresh Liver tissues were collected to detect the expression of TNF-αmRNA and FasmRNA by RT-PCR. Parts of the liver tissues were collected in 10% formaldehyde fixed, then were made in paraffin-embeded specimens to detect rat liver histological change(HE). [Result] The serum levels of ALT in group of EGb were lower than the group of NS significantly (P <0.01) at 2, 6, 24 h after reperfusion. The serum levels of AST in group of EGb were lower than the group of NS significantly (P <0.05) at 2 h, 6 h after reperfusion. The expression of TNF-αmRNA was lower in group of EGb than the group of NS significantly(P <0.05) at 2, 6 h after reperfusion. The expression of FasmRNA was lower in group of Ginkgo Biloba extract than the group of NS significantly (P <0.05) at 2, 6, 24 h after reperfusion. [Conclusion] Extract of Ginkgo Biloba leaves preconditioncan attenuates liver graft ischemia/reperfusion injury and affects the expression of TNF-αmRNA and FasmRNA in the liver graft.
