沙棘汁对铅所致小鼠脑组织生物化学指标的影响

【摘要】 [目的]研究沙棘汁对醋酸铅所致小鼠脑脂质过氧化及胆碱酯酶和单胺类神经递质含量改变的影响.[方法]制作铅中毒小鼠模型,灌胃给予不同剂量的沙棘汁,利用硫代巴比妥酸比色法、碱性羟胺法及荧光法分别测定小鼠脑组织内过氧化脂质、胆碱酯酶及单胺类神经递质含量.[结果]铅组小鼠脑组织内MDA含量明显升高,200,400 g/L沙棘汁降低铅致MDA含量升高,且呈剂量-效应关系;铅组小鼠脑组织内胆碱酯酶活性明显升高,单胺类神经递质中除DA外NE,5-HT,5-HIAA含量均明显下降,400 g/L沙棘汁抑制铅致脑组织内多种神经递质含量的变化.[结论]沙棘汁可拮抗铅所致小鼠脑组织内脂质过氧化及胆碱酯酶和单胺类神经递质含量的改变.

【Abstract】 OBJECTIVE To study the effects of the Hippophae Rhamnoides L juice on lead-induced change of lipid peroxidation,acetylcholinesterase(AchE) and catecholamine neurotransmitters in mice brain. METHODS Mice were administrated peritoneal injection with lead acetate,then mice were given various dosages of Hippophae Rhamnoides L juice by gavage.The value of MDA was determined by the thiobarituric acid colorimetric method,and the activity of AchE was determined by the alkaline hydroxylamine method,and(contents) of NE,DA,5-HT were determined by the flourimetric method. RESULTS The MDA levels were increased in lead-treated mice, which were reduced by 200,400 g/L Hippophae Rhamnoides L in dose-dependent manner.The activities of AchE was significantly increased in the lead-treated group,which were decreased by 400 g/L Hippophae Rhamnoides L.The levels of NE,5-HT,5-HIAA were significantly decreased in the lead-treated mice,and this decreases were antagonized by 400 g/L Hippophae Rhamnoides L,except for that in dopamine. CONCLUSION Hippophae Rhamnoides L juice has protective effects against the lead acetate-induced changes in neurobiochemical parameters.
