霍乱毒素亚单位B结合辣根过氧化酶电镜观察触液神经元几种显色方法比较
Comparative study on staining methods of CB-HRP for visualization of CSF-contacting neurons in electron microscopy
【摘要】 目的寻找电镜观察霍乱毒素亚单位B结合辣根过氧化酶(CB-HRP)最理想的显色方法。方法将CB-HRP引入侧脑室,分别使用二氨基联苯胺(DAB)法、CoCl2-DAB法、TMB-ST法、TMB-ST结合DAB-CoCl2法显色CB-HRP,电镜下观察CB-HRP标记阳性神经元显色情况。结果DAB法、CoCl2-DAB法以及TMB-ST法显色后,电镜下均未见有CB-HRP标记阳性神经元;TMB-ST结合DAB-CoCl2法显色,见有大量CB-HRP标记阳性神经元,且超微结构清晰,标记物明显。结论TMB-ST结合DAB-CoCl2法是用于电镜观察CB-HRP的最理想方法。
【Abstract】 Objective To find the ideal method to stain the CB-HRP labeled structures in electron microscopy.Methods CB-HRP was injected into one of the rat lateral ventricles.Varied methods(DAB,CoCl2 plus DAB,TMB-ST and TMB-ST plus DAB-CoCl2-H2O2) were used to reveal the CSF-contacting neurons.Results CSF-contacting neurons were noticed in the samples stained by using the TMB-ST plus DAB-CoCl2-H2O2 method,while the other methods failed to visualize the CSF-contacting neurons.Conclusion TMB-ST plus DAB-CoCl2-H2O2 is the desired method to reveal CB-HRP in electron microscopy.
