高脂血症大鼠血液流变学特性改变的研究

【摘要】 目的研究高脂血症模型大鼠血液流变学特性的改变。方法12只Wistar大鼠随机分为2组,对照组给予基础饲料喂养;高脂组给予高脂饲料(83%基础饲料,5%蛋黄粉,2%胆固醇,10%猪油)喂养。16周后,腹主动脉采血,检测血浆甘油三酯、胆固醇含量、红细胞膜胆固醇与磷脂的比值、红细胞变形指数、积分变形指数、取向指数电泳率、聚集指数、全血粘度以及血浆粘度。结果通过饮食诱导所获得的高脂血症模型大鼠,其体重、血浆甘油三酯、胆固醇明显高于对照组(P<0.05,P<0.01,P<0.01);红细胞的变形指数、取向指数、电泳率以及红细胞膜的流动性都显著降低(P<0.01,P<0.05,P<0.01,P<0.05);血浆粘度、全血粘度、红细胞聚集指数和红细胞膜胆固醇与磷脂比值显著增高(P<0.01)。结论血脂水平升高可影响血液流变学特性以及红细胞微观流变学特性。

【Abstract】 Objective To explore the changes of hemorheological characteristics and possible mechanisms in rats with hyperlipidemia.Methods Twelve rats were randomly divided into two groups:The control group animals were fed with standard laboratory chow;the hyperlipidenia ones were fed with high-fat diet(standard laboratory chow83%,cholesterol2%,lard 10% and powder of vitellus 5%).Sixteen weeks later,blood was drawn by abdominal aortic puncture,anti-coagulated with heparin;and plasma cholesterol and triglyceride concentration,whole blood viscosity,plasma viscosity,RBCs hemorheological indexes,RBCs electrophoretic mobility,membrane fluidity as well as the value of Ch(cholesterol)/Pl(phospholipid) of erythrocyte membrane were measured.Results The results showed that deformation index,integrated deformation index,orientation index,erythrocyte electrophoretic mobility and membrane fluidity decreased significantly(P<0.01,P<0.01,P<0.05,P<0.01,P<0.05);plasma cholesterol and triglyceride concentration(P<0.01),the whole blood viscosity(P<0.01),plasma viscosity,aggregation index and the value of Ch/Pl increased significantly(P<0.01).Conclusion The rise of plasma lipid can influence hemorheological and erythrocyte microrheological characteristics.
