基质金属蛋白酶活性与新生鼠窒息后心肌损害的相关性研究

【摘要】 目的探讨基质金属蛋白酶3、9(MMP-3、MMP-9)与新生鼠窒息后心肌损害的相关关系。方法建立新生鼠常压窒息模型,观察心肌组织病理改变、免疫组化法测定心肌组织MMP-3、 MMP-9的活性及氯胺T法测定心肌胶原含量。结果窒息后心肌组织MMP-3、MMP-9的活性表达较对照组明显增高(P均<0.05),MMP-3活性以7 d时为最高,MMP-9的活性则逐渐增高。窒息组心肌胶原含量于7、14 d逐渐增高(P均<0.05)。MMP-3、MMP-9的表达与心肌病理积分、胶原含量均呈正相关(P均<0.05)。结论新生鼠窒息后心肌组织MMP-3和MMP-9被激活,与心肌病理损害密切相关,继发胶原含量增高,可能是导致心肌间质的重构、影响心功能的因素之一。

【Abstract】 Objective To explore the relationship between myocardial matrix metalloproteinases (MMP-3,9) activities and cardiac injury in asphyxiated newborn rats. Methods Asphyxia animal model was set, and myocardial histopathological study was done. The MMP-3、9 activities were measured by immunohistochemical assay and myocardial collagen quantity was measured by means of chloramines T. Results In asphyxia group,the activities of MMP-3 and MMP-9 were higher than those of control group (P<0.05). MMP-3 activity reached the highest on day 7,and MMP-9 activity increased gradually. The myocardial collagen content in asphyxia group at 7 d and 14 d were higher than those in control group (P<0.05). MMP-3 and MMP-9 activities positively correlated with myocardial histopathological scores (r=0.669, 0.667. P<0.01) and collagen content (r=0.482,0.679,P<0.05). Conclusions MMPs activities in asphyxiated newborn rats are activated and intimately associated with the cardiac damage. Increased myocardial collagen predicts that MMPs may lead to myocardial interstitial up regulation and affect cardiac function.
