非洲菊不同组织总RNA提取

【摘要】 以非洲菊为材料,采用Trizol试剂法、改良的异硫氰酸胍法、CTAB法和尿素—氯化锂法,提取非洲菊的花、花蕾、叶、花梗和根的总RNA。所得RNA经1%琼脂糖凝胶电泳以及紫外分光光度计分析,结果表明Trizol法和改良的异硫氰酸胍法适用于花和根组织,尿素-氯化锂法对花蕾和叶组织RNA有较好的提取效果,而花梗组织RNA最好采用CTAB法。

【Abstract】 This experiment had used Trizol method,improved Isothiocyanate method,CTAB method and urea-LiCl method to extract total RNA from different tissue of Gerbera jamesonii Bolu.The obtained RNA was analyzed through ultraviolet spectrophotometer and 1 % agarose gel electrophoresis.The results showed that Trizol method and improved Isothiocyanate method fit for extraction RNA from flower and root,urea-LiCl method has better effect for bud and leaf,and CTAB method was useful for the RNA from the pedicel.
