金银花中绿原酸的分离与测定方法

【摘要】 用正交试验法对金银花中绿原酸分离的醇提工艺进行优化,用紫外分光光度法测定了金银花提取物中绿原酸的含量,利用薄层色谱,对不同的展开剂进行了比较筛选,找出了一种能较好的分离出绿原酸的展开剂.从正交试验中得出醇提的最佳工艺:乙醇浓度75%,溶剂量为10倍,提取时间为2小时,提取次数为2次.利用薄层色谱法,用硅胶H,0.7%的CMC板,乙酸丁脂:甲酸:水:乙醇(1:1:1:0.2)上层液为展开剂能较好的分离出金银花提取物中的绿原酸.

【Abstract】 Orthodox-design method Is adopted to research the ethanol-extraction process for chlorogenic acid in lonicerae flower and UV-vis is applied to measure the content of chlorogenic acid in the extraction products.Thin Layer Chromtography(TLC) is undertaken to test the optimum separating system for chlorogenic acid.The optimum process for ethanol-extraction: 10 times(M/M) of 75% ethanol is added to the raw material and extracted for 2 hs.This process must be undertaken twice and the extraction products are put together.The optimum TLC separating system: thin layer plate is adopted with silicon-gel H in 0.75% CMC,solvent is the upper layer of CH3COOC4H9: HCOOH : H20 : CH3CH2OH =1:1:1:0.2.
