血管紧张素Ⅱ对精子运动参数和胞内钙离子的影响
Effect of AngiotensinⅡ on Human Sperm Motility and Intracellular Calcium
【摘要】 目的:探讨血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)体外对不育症精子运动参数和胞内钙离子的影响。方法:不同浓度的AngⅡ在体外与经上游优化处理的不育症精子分别共同孵育20、40min,采用计算机辅助精子分析(CASA)检测人精子运动参数;荧光分光光度计检测胞内游离Ca2+荧光强度的变化。结果:AngⅡ(10nmol/L和100nmol/L)在体外能显著提高不育症精子活率及运动参数(p<0·05),并激发胞内Ca2+快速升高(p<0·001);Losartan(AngⅡ受体AT1拮抗剂)、EGTA可以明显阻断和抑制这些过程。结论:AngⅡ在体外可以显著增高不育症患者精子的运动参数,其结果可能是通过人精子膜上的AT1受体结合作用后使胞外Ca2+内流而实现的。
【Abstract】 s]Objective:To investigate the effects of angiotensinⅡ(AngⅡ) on human sperm motility and intracellular calcium in vitro. Methods:Sperms were aseptically obtained by masturbation and prepared by swim-up technique from 32 infertile males, and then were incubated with different concentrations of AngⅡ at 37℃ in vitro for 20 and 40 minutes respectively. Motility parameters were measured with computer-aided sperm analysis (CASA) and fluorescence intensity of intracellular Ca~ 2+ was measured by fluorospectrophotometer. Results:The results showed that AngⅡ (10nmol/L or 100nmol/L) could significantly increase the motility parameters of male infertility spermatozoa incubated in vitro (p<0.05) and stimulate the increase of the intracellular Ca~ 2+ concentration(p<0.001). However ,these effects of AngⅡ could be inhibited by Losartan(a AngⅡblocker) and EGTA. Conclusion:AngⅡ (10nmol/L or 100nmol/L)can significantly increase the mobility of male infertility sperm in vitro. The mechanism may be explained by the influx of extracellular calcium caused by the integration of Ang II with AT1 on human spermatozoa.
