西瓜细菌性果斑病菌Taq Man探针实时荧光PCR检测鉴定方法的建立
Establishment of Detection and Identification of Acidovorax Avenae Subsp. Citrulli with Taq Man Probe Real-time Fluorescent PCR Method
【摘要】 根据西瓜细菌性果斑病菌(Acidovorax avenaesubsp.citrulli)与相关细菌16S rDNA序列差异,设计出对西瓜细菌性果斑病菌具有稳定点突变特异性探针Aac-probe,利用该探针对23种供试菌株进行了实时荧光PCR检测实验。结果表明,只有西瓜细菌性果斑病菌能检测到荧光增强信号,其它细菌无荧光增强信号。该方法特异性强,灵敏度高,重复性好,可有效地应用于西瓜细菌性果斑病菌的检测。
【Abstract】 A specific probe was designed based on difference of 16S rDNA between Acidovorax avenae subsp.citrulli and other bacterium,and it was used to detect 23 testing strains.The results showed that no signal was detected for the others except Acidovorax avenae subsp.citrulli.The method is more specific than normal PCR,and it was also sensitive method to detect Acidovorax avenae subsp.citrulli.
