在流动血液中蜂胶黄酮对血小板活性的抑制作用

【摘要】 目录:观察蜂胶黄酮对流动状态下血小板活性的影响。方法:实验于2005-12/2006-09在泰山医学院生命科学研究所完成。采用体外灌注法复制血管内膜创伤模型,正常血液流经受损膜表面后,在切应力为1000s-1时,测量血小板黏附率。以0.1g/L的黄酮磷酸盐溶液作实验组,加入25mmol/L的磷酸盐溶液作阴性对照组,同浓度的阿魏酸磷酸盐溶液作阳性对照组。结果:①血小板在基质膜表面的黏附面积百分比:实验组血小板黏附率低于阴性对照组[(10.498±2.548)%,(18.792±4.169)%,F=34.72,P<0.01];而实验组与阳性对照组[(9.933±2.647)%]比较,差异不明显(P>0.05)。②血小板在基质膜表面的黏附状况:当血液流经盖玻片后,可观察到血小板黏附于受损膜表面。阴性对照组血小板分布密度较大,有聚集倾向。实验组血小板分布稀疏,密度较均匀。结论:蜂胶黄酮能抑制血小板的活化,降低其在受损膜表面的黏附活性。

【Abstract】 AIM: To investigate the effects of flavone from propolis on blood-platelet activity in vitro flowing blood. METHODS: The experiment was conducted at the Institute of Life Science, Tanshan Medical College from December 2005 to September 2006. Intimal injury model was made by vitro perfusion. Blood-platelet adhesive rate was detected when blood went through surface in 1 000s-1 shear rate. Three groups were divided: experimental group (0.1 g/L, flavone from propolis), negative control group (25 mmol/L, phosphate solution) and positive control group (0.1 g/L, ferulic acid). RESULTS: ①Percentage of adhesive area on matrix membrane surface of plaque: The platelet adhesion on area of experimental group was lower than that of negative control group (10.498±2.548)%,(18.792 ±4.169)%,F=34.72,P < 0.01 . There was no significant difference between experimental group and positive control group (9.933±2.647)%, P > 0.05 . ②Adhesion of platelet in matrix surface: Platelet adhered to the injured membrane when blood flowed on coverslip. In the negative control group, the plaque distributed densely, with gathered trend. In the experimental group, plaque scattered evenly. CONCLUSION: Flavone from propolis can inhibit platelet activation and decrease its adhesion on injured surface.
