解郁1号影响抑郁大鼠皮质及海马区脑源性神经营养因子蛋白表达的效应

【摘要】 目的:观察以调理脾胃法组成的中药复方解郁1号对抑郁大鼠皮质、海马区脑源性神经营养因子蛋白表达的影响。方法:实验于2004-03/04在广西中医学院中心实验室完成。SD大鼠40只,按随机表随机分为正常组、模型组、阿米替林组、解郁1号12,6g/(kg·d)剂量组,每组各8只。解郁1号为半夏,竹茹,枳实,茯苓,柴胡,石菖蒲,合欢花各10g等组成,中药购于广西中医学院附属一院中药房,经中药植化室鉴定后,水煎浓缩为1.5g/mL,置4℃冰箱内备用。采用慢性不可预见性应激诱导抑郁大鼠模型,各组在造模同时即开始给药,分别胃饲阿米替林10mg/(kg·d)(蒸馏水配置,浓度为1g/L),解郁1号提取液12,6g/(kg·d),模型组、正常组胃饲生理盐水(6mL/kg),每天8:00开始,除正常组外,各组接受21d长期慢性刺激。各组动物末次造模结束后,采用旷场实验进行行为学评分,24h后,立即断头,迅速冰皿上取脑,脑组织放入液氮罐内速冻5min,取出后冰冻切片机进行冰冻切片,片厚20μm,-20℃冰箱保存,用免疫组织化学法进行大鼠皮质、海马CA4内脑源性神经营养因子蛋白表达检测。结果:实验大鼠38只进入结果分析。①与正常组比较,模型组额叶皮质、海马CA4区脑源性神经营养因子免疫标记阳性神经元数目明显减少(257.00±38.40,244.4±38.82,P<0.01),而且免疫标记着色较浅,部分神经元胞浆有空化现象。②与模型组比较,阿米替林组、解郁1号12,6g/(kg·d)剂量组均能增强脑源性神经营养因子在额叶皮质、海马CA4区的表达(皮质神经元个数:394.25±27.70,403.5±27.77,378.38±32.61;海马CA4区神经元个数:363.29±29.94,354.50±41.07,345.00±36.39,P<0.01),额叶皮质、海马CA4区脑源性神经营养因子免疫标记阳性神经元形态未见明显异常。结论:解郁1号具有抗抑郁作用,其作用机制可能通过增强脑源性神经营养因子蛋白表达,从而发挥抗神经元损伤作用,达到抗抑郁目的。

【Abstract】 AIM: To investigate the effect of No.1 Jieyu decoction (Jieyu decoction), which is a complex prescription with the function of adjusting the spleen and stomach, on brain-derived neurotrophic factor (BDNF) protein expression in the cortex and hippocampus of depression rats. METHODS: The experiment was conducted in the Central Laboratory of Guangxi TCM College from March to April 2004. Fourty SD rats were selected and randomly divided into normal control group, model group, amitriptyline group (AT group) and 12 g/(kg·d) as well as 6 g/(kg·d) Jieyu groups with 8 rats in each group. No.1 Jieyu decoction was prepared by 10 g of pinelliae tuber, caulis bambusae in taenisam, orange pease, hoelen, Chinese thorowax root, grassleaf sweetflag rhizome and flos albiziae respectively, and the traditional Chinese medicine was bought from the TCM dispensary of First Hospital Affiliated to Guangxi TCM College, which was identified by the Department of TCM Phytochemistry and decocted in water to obtain the concentrated decoction (1.5 g/mL) and then placed in the refrigerator at 4 ℃ for using. Rat models of chronic unpredictable stress induced depression were adopted, and the administration was given at the same time of modeling. Rats in all groups were given gastric perfusion of AT at 10 mg/(kg·d), which was prepared by distilled water at the concentration of 1 g/L, Jieyu extraction of 12 g/(kg·d) and 6 g/(kg·d), while rats in the model group and normal control group were administrated with normal saline at 6 mL/kg. The administration was given at 8.00 PM, and rats, except those in the normal control group, were given a 21-day chronic long-term stress. Rats were praxiologically evaluated by open field test after the last modeling, and executed at 24 hours later to obtain the brain as soon as possible on the ice-plate. The brain tissues were frozen for 5 minutes in the nitrogen canister and then taken out to cut into frozen pieces at the thickness of 20 μm and put into the refrigerator at -20 ℃. Immunohistochemical method was used to determine the BDNF protein expression in the cortex and CA4 hippocampus of rats. RESULTS: A total of 38 experimental rats were involved in the analysis of results. ① The positive neurons immune-labeled by the BDNF in cortex of frontal lobe and hippocampal CA4 region were significantly less in the model group than those in the normal control group (257.00±38.40, 244.4±38.82, P < 0.01), while the immune label was in light color and cavitation was found in the cytoplasm of partial neurons. ② Compared with the model group, the BDNF expressions in the cortex of frontal lobe and hippocampal CA4 region were reinforced in the AT group, 12 g/(kg·d) and 6 g/(kg·d) Jieyu decoction groups (The number of cortical neurons: 394.25±27.70, 403.5±27.77, 378.38±32.61; number of neurons in hippocampal CA4 region: 363.29±29.94, 354.50±41.07, 345.00±36.39, P < 0.01). However, the shapes of positive immune-labeled neurons of BDNF in the cortex of frontal lobe and hippocampal CA4 region were not significantly changed. CONCLUSION: Jieyu decoction can inhibit the occurrence of depression, and its mechanism of action may suppress the injury in neurons by improving the BDNF protein expressions, and thus suppress the depression.
