脑卒中后抑郁大鼠模型的建立

【摘要】 目的:探索制备理想的脑卒中后抑郁大鼠模型的方法,为研究脑卒中后抑郁的中西医药物治疗提供一个良好的动物模型。方法:实验于2002-07/2003-02在成都中医药大学实验室完成。SD大鼠45只随机分为正常对照组10只、单纯脑卒中组10只、孤养应激组10只、脑卒中后抑郁组15只。采用线栓法制备局灶性脑缺血大鼠模型,在此基础上综合孤养、中度的不可预测应激处理复合制备脑卒中后抑郁大鼠模型。运用Longa5评分法(评分越高,神经功能缺损越严重)和水平木棒法(正常大鼠放置其上可以在上面持续时间超过3min)评定脑卒中大鼠神经功能恢复程度。运用糖水实验、敞箱实验、被动躲避实验评定脑卒中后抑郁大鼠的兴趣感变化、自发活动和探究行为等抑郁行为学改变。采用荧光分光度法测定大鼠脑内单胺类神经递质(去甲肾上腺素、5-羟色胺、多巴胺)水平。结果:①在造模中6只大鼠死亡,3只大鼠造模未成功,36只大鼠进入结果分析。②脑卒中大鼠22只(包括单纯脑卒中组9只,脑卒中后抑郁组13只),清醒后第4,8,24小时的神经功能缺损评分分别为2.58±0.69,2.32±0.58,1.37±0.60。③脑卒中动物20只(包括单纯脑卒中组8只,脑卒中后抑郁组12只),脑卒中后第1,3,5天在圆棒上停留时间分别为(110.94±31.40)s,(149.53±16.56)s,(169.88±8.44)s。④脑卒中后抑郁组脑卒中后第7,14天的体质量显著低于正常对照组[(348.8±47.7)g,(390.9±22.9)g,P<0.05;(321.7±43.8)g,(392.6±23.5)g,P<0.01]。⑤脑卒中后抑郁组糖水消耗量显著低于正常对照组[(8.48±1.15)mL/kg,(113.0±11.8)mL/kg,P<0.01]。⑥脑卒中后抑郁大鼠敞箱活动低下,被动躲避缺陷。⑦脑卒中后抑郁组双侧额顶皮质及脑干的去甲肾上腺素、5-羟色胺显著降低。结论:在局灶性脑缺血大鼠模型的基础上复合孤养和应激处理可以制备出较理想的脑卒中后抑郁大鼠模型。

【Abstract】 AIM:To explore the method to set up an ideal rat model of poststroke depression,provide a good animal model for Chinese and western medicine care.METHODS:The experiment was conducted from July 2002 to February 2003 at the Laboratory of Chengdu University of TCM.The 45 SD rats were randomly divided into normal control group with 10 rats,simple stroke group with 10 rats,stress group with 10 rats and poststroke depression group with 15 rats.Focal cerebral ischemic rat models were made with thread embolization method.Poststroke depression rat models were established with comprehensive separately breeding and moderate unpredictable stress on this basis.The neurological functional recovery of stroke rat was assessed by Longa 5 grades(the higher score represented severe neurological functional impairment)and horizontal bar test(lasting more than 3 minutes if normal rats were put on it).Sucrose solution consumption test,open-field test and passive avoid test were used to estimate the change of interest,the level of spontaneous motor activities and investigative behavior of poststroke depression rat models.Levels of monoamine neurotransmitters(noradrenaline,5-serotonin,dopamine)in brain tissue of rats were measured with fluorescent spectrophotometric method.RESULTS:①Six rats died in the model building,and 3 rats were failed in model building,so 36 rats were involved in the result analysis.②The neurologic impairment scores were(2.58±0.69,2.32±0.58,1.37±0.60),respectively at 4,8 and 24 hours after awaking in 22 stroke rats(including 9 rats in the simple stroke group and 13 rats in poststroke depression group).③Duration in round rod was(110.94±31.40)s,(149.53±16.56)s,(169.88 ±8.44)s at days 1,3 and 5 after stroke in the 20 stroke rats(8 rats in the simple stroke group,12 rats in the poststroke depression group).④The body mass was significantly lower in the poststroke depression group than in the normal control group at days 7 and 14 [(348.8±47.7)g,(390.9±22.9)g,P < 0.05;(321.7±43.8)g,(392.6±23.5)g,P < 0.01].⑤Glucose consumption in the poststroke depression group was lower obviously than that in the normal control group [(8.48±1.15)mL/kg,(113.0±11.8)mL/kg,P < 0.01].⑥The rats in the in the poststroke depression group were low open-field activity and passive avoid impairment.⑦The noradrenaline and 5-serotonin reduced markedly in the bilateral forehead cortex and brain stem in the poststroke depression group.CONCLUSION:On the basis of focal cerebral ischemic rat models,separately raised condition and stress disposal are applied successively to set up suitable poststroke depression rat models.
