马齿苋乙醇提取物对小鼠常压耐缺氧能力的影响

【摘要】 目的:观察马齿苋乙醇提取物对小鼠常压耐缺氧作用的影响,分析马齿苋的药用价值。方法:实验于2005-11/12在延边大学基础医学院机能学实验中心完成。①马齿苋乙醇提取物制备:称取马齿苋干草600g粉碎,用体积分数为0.8的乙醇在水浴中回流提取2次,各用10倍量与8倍量乙醇提取2h和1h,过滤,合并滤液浓缩,干燥成粉末,临用前配制成所需浓度。②取43只健康雌雄昆明种小鼠按随机表法分为5组:空白对照组9只、阳性对照药组8只和3个不同剂量的马齿苋乙醇提取物组(0.25g/kg组9只,0.5g/kg组9只,1g/kg组8只),分别灌胃给予蒸馏水、地奥心血康(0.19g/kg)和马齿苋乙醇提取物0.25,0.5,1g/kg。1次/d,连续给药10d。③观察小鼠的缺氧惊厥时间、缺氧存活时间及耗氧量的变化。结果:43只小鼠进入结果分析。①各组小鼠缺氧惊厥时间和存活时间:与空白组比较,马齿苋0.5g/kg组能明显延长小鼠缺氧惊厥时间(13.03±2.04),(15.30±1.55)min,P<0.05);与空白组比较,阳性对照药组和马齿苋0.5,1g/kg组,能明显延长小鼠缺氧存活时间(13.85±1.89),(15.77±1.75),(16.05±1.65),(15.77.±1.75)min,P<0.05)。②各组小鼠死亡时的总耗氧量及不同时间的小鼠存活率:与空白组比较,阳性对照药组和马齿苋乙醇提取物0.25,0.5,1g/kg组能增加缺氧小鼠在15min时的存活率(33%,63%,44%,67%,75%);,阳性对照药组和不同剂量马齿苋组乙醇提取物组小鼠存活期总耗氧量均无明显改变。③各组小鼠各时间段耗氧量及不同时间的累计耗氧量:与空白组比较,阳性对照药组和马齿苋乙醇提取物组0.5g/kg组能明显降低前5min小鼠的耗氧量(10.16±0.75),(9.09±0.92),(9.30±0.67)mL,P<0.05),而对10min和15min的累计耗氧量无明显影响。阳性组和马齿苋乙醇提取物组0.5g/kg组能明显降低0~5min时间段的耗氧量,而5~10min和10~15min时间段的耗氧量则高于空白组。结论:马齿苋乙醇提取物可以明显提高小鼠的常压耐缺氧能力。

【Abstract】 AIM: To study the effects of ethanol extract from Portulaca oleracea L. on hypoxia tolerance of mice, and evaluate the medicinal value. METHODS: The experiment was conducted in the Experiment Center for Function, Basic Medical Science College of Yanbian University from November to December 2005. ①Preparation of ethanol extract from Portulaca oleracea L: 600 g dry Portulaca oleracea L. was grinded and extracted from water bath containing 0.8 volume fraction of alcohol for twice following by extraction for 2 hours and 1 hour respectively by 10 times and 8 times alcohol. After filtration, the filter liquor was concentrated and dried. The proper concentration was prepared. ②Forty-three healthy Kunming mice either male or female were randomly divided into 5 groups: control group (n=9), positive control medicine group (n=8) and three alcohol extracts groups with different doses [0.25 g/kg group (n=9), 0.5 g/kg group (n=9) and 1 g/kg (n=8)]. The mice were intragastrically infused respectively with distilled water, 0.19 g/kg Di’ao Xinxuekang and 0.25, 0.5 and 1.0 g/kg ethanol extract, once daily for 10 days. ③The changes of hypoxia eclamptic time, survival time and oxygen consumption of mice were measured. RESULTS: All the 43 mice were involved in the result analysis. ①Eclamptic time and survival time of mice in each group: Compared with the control group, the eclamptic time of the 0.5 g/kg extract group was longer [(13.03±2.04), (15.30±1.55) minutes, P < 0.05]; compared with the control group, the survival time of the positive control group and 0.5 g/kg extract group was longer [(13.85±1.89), (15.77±1.75), (16.05±1.65), (15.77±1.75) minutes, P < 0.05]. ②The total oxygen consumption when the mice died and the survival time of mice at different time points: Compared with the control group, the positive control group and the ethanol extract groups at the dose of 0.25, 0.5 and 1 g/kg could increase the survival time of mice with hypoxia for 15 minutes (33%, 63%, 44%, 67%, 75%); no change in total oxygen consumption was found in the survival time of mice in the positive control group and the ethanol extract groups at different doses. ③Oxygen consumption at each time point and cumulative oxygen consumption at different time points: Compared with the control group, the positive control group and the ethanol extract groups at the dose of 0.5 g/kg could significantly decrease the oxygen consumption of mice with hypoxia for 15 minutes [(10.16±0.75), (9.09±0.92), (9.30±0.67) mL, P < 0.05], but could not change the cumulative oxygen consumption of mice with hypoxia for 10 and 15 minutes. The positive control group and the ethanol extract groups at the dose of 0.5 g/kg could significantly decrease the oxygen consumption of mice with hypoxia for 0-5 minutes, but the oxygen consumption in 5-10 and 10-15 minutes was higher than the control group. CONCLUSION: Ethanol extract from Portulaca oleracea L. can obviously increase the ability of hypoxia tolerance of mice.
