头穴针刺对急性脑出血大鼠痛反应神经元电活动的双向调节

【摘要】 目的:采用微电极技术,观察大鼠脑出血前后头穴针刺对其甩尾反射潜伏期和神经元放电变化的双向影响。方法:实验于2000-05/2001-03在哈尔滨医科大学电生理实验室完成。①体质量225~250g雄性Wistar大鼠32只。按随机数字法分为4组:正常组,盐水组,注血组,针刺组,每组8只。②注血组和针刺组大鼠采用自身动脉血注入法,复制Wistar大鼠急性脑出血模型后,针刺组大鼠头穴透刺同时采用微电极技术,电麻仪为G-6805型,正极接百会,负极接太阳。选参数为1V,15Hz,通电时间为10min。盐水组注入等量生理盐水,其余步骤同注血组。③采用辐射热照尾诱发放电的方法,观察大鼠脑出血前后头穴针刺对甩尾反射潜伏期和痛反应神经元放电变化。结果:①左尾核及左束旁核注血组在注血后各时间窗内痛兴奋神经元放电与注血前相比潜伏期延长,净增值减少(P<0.05,P<0.01),痛抑制神经元放电与注血前相比抑制时程延长,净增值减少(P<0.01),并有随时间的延长而加重的趋势。②左尾核及左束旁核针刺组痛兴奋神经元在注血前针刺后潜伏期延长,净增值减少(P<0.01);痛抑制神经元在注血前针刺后抑制时程缩短,净增值增加(P<0.01)。③针刺组在注血后针刺后与注血组比较均有极显著性差异(P<0.01),使延长的潜伏期和抑制时程缩短,减少的净增值增加。结论:急性脑出血大鼠痛反应神经元的电活动在注血早期即受到抑制,并有随时间的推移而逐渐加重的趋势。针刺对脑出血前后痛反应神经元的电活动具有双向良性调整作用。

【Abstract】 AIM: To observe the effect of scalp acupuncture to latency of tail-flick and electric discharging of pain-reflection neuron of rats after cerebral hemorrhage by microelectrode technique. METHODS: The experiment was conducted in the Electrophysiological Laboratory of Harbin Medical University between May 2000 and March 2001. ①Thirty-two male Wistar rats (225-250 g) were selected and randomly divided into 4 groups: normal group, physiological saline group, blood-injecting group, and scalp acupuncture group with 8 rats in each group. ②With the injection of blood, the models of acute cerebral hemorrhage were established in the rats of the blood-injection and scalp acupuncture groups. Besides the rats in the scalp acupuncture group were acupunctured with electroacupuncture, the microelectrode technique was adopted with G-6805 type electric acupuncture apparatus. The positive electrode was connected with Baihui and the negative electrode with Taiyang with 1 V, 15 Hz for 10 minutes. The rats of the physiological saline group were injected with matching physiological saline, and the other steps were the same to the blood-injecting group. ③The latency of tail-flick and electric discharging of pain-reflection neuron of rats were observed with the radiation-heat tail irradiating method before and after cerebral hemorrhage. RESULTS: ①The latency of electric discharging of pain-reflection neuron in left caudate nucleus and left nucleus parafascicularis of the blood-injecting group at different periods was longer than that before blood injecting, and the net increment was decreased (P < 0.05, P < 0.01); the inhibitory course of pain-reflection neuron was prolonged compared with that before blood injecting, and the net increment was decreased (P < 0.01), which was prone to be serious as time passed. ②The latency of electric discharging of pain-reflection neuron in left caudate nucleus and left nucleus parafascicularis of the scale acupuncture group was longer than that before blood injecting, and the net increment was decreased (P < 0.01); the inhibitory course of pain-reflection neuron was shortened compared with that before blood injecting, and the net increment was increased (P < 0.01). ③There were significant differences between the scale acupuncture group after blood injecting and acupuncture and the blood-injecting group (P < 0.01), which shortened the prolonged latency and inhibitory time and increased the decreased net increment. CONCLUSION: The electric-activity of neuron of rats after acute cerebral hemorrhage is inhibited just in the early period of injecting blood, and the inhibition will be more serious with the time goes on. Scale acupuncture has good bidirectional adjustment effect on the electric-activity of pain-reflection neuron before and after cerebral hemorrhage.
