亚低温对大鼠缺血脑组织血管内皮生长因子表达的影响
Influence of mild hypothermia on expression of vascular endothelial growth factor after cerebral ischemia in rats
【摘要】 目的:观察亚低温对大鼠脑缺血后血管内皮生长因子的表达的影响,探讨亚低温的脑保护作用机制。方法:实验于2005-05/07在武汉大学人民医院神经内科实验室完成。选择成年雄性SD大鼠20只,随机数字表法分为常温对照组和亚低温组,每组10只。采用改良线栓法制备持续性大脑中动脉闭塞模型,神经功能缺损评分1~3分为模型成功标志。常温对照组在室温下喂养,亚低温组在术后2h开始低温诱导,肛温控制在34~35℃,持续72h,两组均在术后第7天断头处死取脑,测定脑梗死体积和免疫组织化学染色检测血管内皮生长因子的表达(胞浆呈棕黄色为阳性细胞)。结果:纳入动物20只,均进入结果分析。大脑中动脉阻塞后,亚低温组脑梗死体积明显低于常温对照组[分别为(153.25±23.14),(253.45±36.21)mm3,P<0.01]。亚低温组血管内皮生长因子阳性细胞数明显低于常温对照组[分别为(24.02±5.05),(36.07±2.69)个/高倍视野,P<0.01]。结论:亚低温能使脑梗死体积减小,抑制血管内皮生长因子的表达,亚低温抑制血管内皮生长因子表达可能是亚低温脑保护机制之一。
【Abstract】 AIM: To investigate the effect of mild hypothermia on expression of vascular endothelial growth factor (VEGF) after cerebral ischemia in rats and study the cerebral protective mechanism of mild hypothermia. METHODS: The experiment was performed at the Laboratory of Department of Neurology,Renmin Hospital,Wuhan University between May and July 2005. Totally 20 adult male SD rats were divided randomly into normal temperature group and mild hypothermia group with 10 in each group. A permanent middle cerebral artery occlusion (MACO) model was established with modified nylon suture embolization. 1-3 points of neurologic impairment was considered as successful model. The rats of normal temperature group were fed in the room temperature,and the temperature of the rats of mild hypothermia group were controlled at the range 34-35 ℃ after surgery two hours for 72 hours. Seven days later,the rats were decapitated to take out the heads. The infarct volume was measured. The expression (positive cells with buffy kytoplasm) of VEGF was measured with immunohistochemical method. RESULTS: Totally 20 included animals were involved in the result analysis. After MACO,the infarct volume of mild hypothermia group was significantly smaller than that in the normal temperature group (153.25 ±23.14),(253.45±36.21) mm3,respectively,P < 0.01. Compared with the normal temperature group,the expression of VEGF positive cells was fewer in the mild hypothermia group (24.02±5.05),(36.07±2.69) piece/high power field,P < 0.01. CONCLUSION: Mild hypothermia can make the infarct volume decrease and inhibit the expression of VEGF. It may be one of the cerebral protective mechanism of mild hypothermia that mild hypothermia inhibits the expression of VEGF.
