去卵巢大鼠骨质疏松模型的特点
Characteristics of osteoporosis models in ovariectomized rats
【摘要】 目的:观察去卵巢致Wistar大鼠骨质疏松模型骨密度、结构、最大载荷等变化特点,探讨骨量与骨结构的可能关系。方法:实验于2002-09/2004-01在解放军第三军医大学动物研究所及中心实验室完成。选择雌性未孕Wistar大鼠20只,随机数字表法分为去势组、假手术组,各10只。去势组行双侧卵巢切除,假手术组不切除卵巢。术后8周测定以下指标:①骨密度:在双能X射线骨密度仪的探头下,应用小动物软件进行腰椎和股骨的扫描,测定骨密度值。②椎体压缩测试:将磨制好的椎体置于实验机上,取加载速度1mm/min进行测试,其结构力学参数由载荷-变形曲线获得,材料力学参数由应力-应变曲线获得。③骨形态计量学检查指标:骨小梁面积百分数(%Tb.Ar),代表骨量;骨小梁厚度(Tb.Th)、骨小梁数量(Tb.N),骨小梁分离度(Tb.Sp)。④血、尿生化指标的测定:血清钙、磷,尿钙、磷,尿肌酐,均采用BECKMAN自动生化分析仪。血清骨钙素、血清碱性磷酸酶、血清抗酒石酸酸性磷酸酶、尿羟脯氨酸具体参照试剂盒说明。结果:纳入动物20只,均进入结果分析。①去势组大鼠股骨骨密度明显低于假手术组[分别为(0.166±0.026),(0.195±0.027)g/cm2,P<0.05],腰椎骨密度尤其显著[分别为(0.190±0.011),(0.251±0.029)g/cm2,P<0.01]。②去势组大鼠股骨最大载荷与假手术组相比显著降低[分别为(94.43±8.96),(109.49±12.41)N,P<0.05],腰椎最大载荷降低尤其显著[分别为(98.75±8.19),(118.82±15.74)N,P<0.01]。③去势组大鼠骨小梁面积百分数(%Tb.Ar)、骨小梁厚度(Tb.Th)、骨小梁数量(Tb.N)与假手术组相比显著降低(去势组分别为4.78±0.76,46.29±9.96,0.64±0.08,假手术组分别为7.74±1.76,60.91±3.80,1.26±0.26,P<0.01);骨小梁分离度(Tb.Sp)显著增高(分别为2300.85±480.10,1112.27±65.81,P<0.01)。④血、尿生化指标显示:骨吸收大于骨形成。结论:Wistar大鼠去卵巢8周成功建立了绝经后骨质疏松模型,骨量与骨结构的一些参数可能存在某种关系。
【Abstract】 AIM: To observe the characteristics of bone mineral density (BMD), bone structure and maximum load in osteoporosis models of ovariectomized Wistar rats, and explore the possible relationship between BMD and structure. METHODS: The experiment was conducted at the Animal Institute and Central Laboratory, the Third Military Medical University of Chinese PLA from September 2002 to January 2004. Twenty female non-pregnant Wistar rats were randomly divided into sham group and ovariectomy (OVX) group, with 10 rats in each. The rats in OVX group were subjected to excise bilateral OVX, while sham operation was performed in sham group. At the postoperative 8th week, the following markers were measured:①BMD: Lumbar vertebra and femur were scanned under the detecting head of double-functional X ray BMD apparatus to detect BMD values.②Compression test of vertebral body: The milled vertebral bodies were tested at the load speed of 1 mm per minute. The parameter of structural mechanics was expressed as load-deformation curve while the parameter of material mechanics was expressed as stress-strain curve.③Histomorphometric analysis of bone: percentage of trabecular area (%Tb.Ar) on behalf of bone mass; trabecular thickness (Tb.Th), trabecular number (Tb.N), trabecular separation (Tb.Sp).④Biochemical markers of serum and urine: Calcium and phosphorus in serum and urine as well as urine creatinine were all determined with BECKMAN automatic biochemistry analyzer. Osteocalcin, alkaline phosphatase and anti-tartaric acid phosphatase in serum and urinary hydroxyproline were referred to the instruction of Test Kit. RESULTS: Twenty rats were all involved into the result analysis.①Compared with sham group, BMD and maximum load of femur were obviously decreased in OVX group [(0.166±0.026), (0.195±0.027) g/cm2, P < 0.05; (94.43±8.96), (109.49±12.41) N, P < 0.05], especially in lumbar vertebra [(0.190±0.011), (0.251±0.029) g/cm2, P < 0.01; (98.75±8.19), (118.82±15.74) N, P < 0.01].②%Tb.Ar, Tb.Th and Tb.N of rats were remarkably lower in OVX group than in sham group (OVX group: 4.78±0.76, 46.29±9.96, 0.64±0.08; Sham group: 7.74±1.76, 60.91±3.80, 1.26±0.26, P < 0.01); However, Tb.Sp in OVX group elevated significantly (2 300.85 ±480.10, 1 112.27±65.81, P < 0.01).④The biochemical markers of serum and urine showed: Bone resorption was more than bone formation. CONCLUSION: After 8 weeks of ovariectomy, the postmenopausal osteoporosis models are established successfully in Wistar rats. There are some relations between BMD and some bone structure parameters.
