丹参舒颈丸对抗脊髓型颈椎病模型大鼠氧自由基损伤

【摘要】 目的:观察丹参舒颈丸对脊髓型颈椎病模型大鼠血液和损伤区脊髓匀浆内超氧化物歧化酶活性和丙二醛含量的影响。方法:实验于2004-01/12在中国中医研究院骨伤科研究所中药药理试验室完成。①选用健康8周龄清洁级SD雄性大鼠60只。随机抽取10只大鼠为假手术组:只暴露颈椎椎板,不放置硅胶颗粒。其余50只均在不剪除第4,5颈椎情况下,将硅胶颗粒置于其间,造成脊髓型颈椎病模型。根据组间均衡一致的原则将造模后大鼠随机分为5组:丹参舒颈丸高、中、低剂量组、颈复康组和模型组,每组10只。造模后1周开始灌胃给药,共30d(从造模后1周开始)。丹参舒颈丸高、中和低剂量治疗组:灌胃含丹参舒颈丸(主要成分:丹参、葛根、羌活、白术、木瓜、白芍、杜仲、西红花等;批准文号:(2005)京药制字[365]第F-2181号,9g/袋;由北京万和颈椎病医院提供)生药6,3,1.5g/kg溶液;颈复康组:灌胃含颈复康(承德中药集团有限责任公司出品;批准文号:ZZ-5464-翼卫药准字(1995)第080193号,5g/袋)生药1.7g/kg;假手术组和模型组:灌胃等量生理盐水。②干预结束后,采用黄嘌呤氧化法和硫代巴比妥酸化学发光法测定血液超氧化物歧化酶活性和丙二醛水平。按南京建成生物工程研究所提供试剂盒说明,测定脊髓匀浆丙二醛含量及超氧化物歧化酶活性。③计量资料差异比较采用单因素方差分析和Q检验。结果:SD大鼠60只均进入结果分析。①模型组、丹参舒颈丸中剂量组和颈复康组血液超氧化物歧化酶活性明显低于假手术组和丹参舒颈丸低剂量组(P<0.05),模型组血液丙二醛水平明显高于假手术组和丹参舒颈丸低剂量组(P<0.05)。②丹参舒颈丸各剂量组脊髓组织超氧化物歧化酶活性明显高于模型组和假手术组(P<0.05~0.01),模型组和颈复康组明显低于假手术组(P<0.05)。丹参舒颈丸各剂量组脊髓组织丙二醛含量明显低于模型组(P<0.01),假手术组低于其他5组(P<0.05)。结论:丹参舒颈丸可以降低脊髓型颈椎病模型大鼠血和脊髓内丙二醛含量,增加超氧化物歧化酶活性,从而发挥清除体内氧自由基的作用;其中低剂量丹参舒颈丸对血液中丙二醛水平和超氧化物歧化酶活性改善作用明显优于其他剂量和颈复康。

【Abstract】 AIM: To observe the effect of Danshen shujing wan (DSW) on the activity of superoxide dismutase (SOD) and content of malondialdehyde (MDA) in blood and spinal homogenate of rats with cervical spondylotic myelopathy (CSM). METHODS: The experiment was completed at the Pharmacological Laboratory, Institute of Orthopedic and Traumatology, China Academy of Traditional Chinese Medicine between January and December 2004.①Sixty healthy male SD rats of 8 weeks old and cleaning grade were randomly divided into 6 groups: pseudo-operative group (N group), model group (M group), 3 DSW groups of high, medium and low dosages, and group treated with Jingfukang (J group), with 10 rats in each. The rats in N group were only exposed of cervical vertebral lamina without silica gel, while the rats of other groups were prepared for CSM models by placing silica gel. From 7 days after model establishments, the rats were respectively treated with intragastric infusions, totally for 30 days. DSW groups: 6, 3 and 1.5 g/kg solution containing DSW crude drug (Components: dan-shen root, thomson kudzuvine root, forbes notopterygium rhizome, largehead atractylodes rhizome, flowering quince fruit, white peony root and saffron; Approval code: (2005)365 No.F-2181, 9 g a pocket; Provided by Beijing Wanhe Hospital of Cervical Syndrome); J group: 1.7 g/kg solution containing Jingfukang crude drug (produced by Chengde Traditional Chinese Medicine Group Corporation; Approval code: ZZ-5464-(1995) No.080193, 5 g per pocket); N group and M group: Equal saline.②One month after the treatment, the activity of SOD and content of MDA in blood were detected with xanthine oxidase (XO) and thiobarbituric acid (TBA) chemiluminescence. Referring to the kit instruction offered by Nanjing Jiancheng Biological Engineering Institute, the activity of SOD and content of MDA in spinal homogenate were measured.③The date was statistically analyzed with one-way analysis of variance and Q-test. RESULTS: Sixty SD rats were involved in the result analysis.①Activity of SOD in blood was obviously lower in M group, DSW group of medium dosage and J group than in N group and DSW group of low dosage (P < 0.05), and the content of MDA in blood was obviously higher in M group than in N group and DSW group of low dosage (P < 0.05).②Activity of SOD in spinal tissue was markedly higher in DSW groups than in M group and N group (P < 0.05-0.01), but markedly lower in M group and J group than in N group (P < 0.05). In addition, the content of MDA in spinal tissue was significantly lower in DSW groups than in M group (P < 0.01), and lower in N group than other 5 groups (P < 0.05). CONCLUSION: It is effective to reduce content of MDA and increase activity of SOD in the blood and spinal tissue of CSM rats by administrating DSW, so as to eliminate the effect of oxygen-derived free radical in vivo; Moreover, DSW at low dosage can obviously improve the activity of SOD and content of MDA in blood, compared with DSW at high and medium dosages and Jingfukang.
