光氧化反应增加去细胞牛颈静脉的组织稳定性
Photooxidation in the reinforcement of stability of decellular bovine jugular tissues
【摘要】 目的:观察去细胞及去细胞后光氧化反应对牛颈静脉的组织稳定性的影响。方法:实验于2005-06/09在中南大学湘雅二医院胸心外科实验室完成。①将新鲜牛颈静脉纵形剪开成血管片,横行切取牛颈静脉血管片成6根血管条,随机分为6组,新鲜组,去细胞组,及根据不同的光氧化处理时间将去细胞结合光氧化处理组分为去细胞后光氧化反应12,24,36和48h组。②通过测定处理前后的组织厚度、热皱缩温度、抗张强度及组织蛋白提取分析来比较组织稳定性。结果:①各组在处理前后的厚度:去细胞处理后血管片比处理前变薄[(0.36±0.85),(0.43±0.94)mm,P<0.05],而去细胞再光氧化处理后的血管片厚度无明显改变(P>0.05)。②各组血管片组织稳定性评价:去细胞组热皱缩温度及抗张强度比新鲜组、去细胞后光氧化反应12h组、24h组、36h组、48h组明显降低[热皱缩温度:(69.75±0.54),(72.50±0.53),(73.80±0.59),(74.40±0.46),(75.10±0.21),(75.20±0.26)℃,P<0.05;抗张强度:(3.13±0.94),(5.19±0.65),(4.54±0.88),(4.67±0.60),(5.55±0.43),(5.80±0.82)MPa,P<0.05];经光氧化处理后热皱缩温度上升且高于新鲜组(P<0.05);而最大应力与新鲜组比较无明显变化(P>0.05);去细胞后光氧化反应组的组织蛋白提取量明显少于新鲜组及去细胞组,且随光氧化反应时间延长,组织蛋白提取量逐步减少。结论:去细胞处理降低了牛颈静脉的组织稳定性,而经光氧化进一步处理后组织稳定性增强,且随光氧化时间延长组织稳定性增强。
【Abstract】 AIM To observe effect of photooxidation on stability of bovine jugular tissues of decellular and after decellular. METHODS The experiment was conducted in the Laboratory of Department of Cardiothoracic SurgeryXiangya Second Hospital of Central South University from June to September 2005.①Fresh bovine jugular conduits were length-wisely cut into vascular slices and then crossly cut into 6 vascular strips which were randomly divided into 6 groups including fresh group decellular group and 12 hour group 24 hour group 36 hour group 48 hour group according to different time of decellular bovine jugular crosslinked with photooxidation group.②Tissue stability were compared with determination of tissue thickness heat shrinking temperature tensile strength TS and tissue protein extraction assay. RESULTS ①Thickness before and after treatment in each group Vascular slices of decellular after treatment were thinner than those before treatment (0.36±0.85),(0.43±0.94) mm,P < 0.05 while there were no obvious changes in thickness of decellular vascular slices after re-photooxidation P > 0.05.②Evaluation on stability of tissues in vascular slices of each group Heat shrinking temperature and TS in the decellular group were remarkably decreased than those in other groups Heat shrinking temperature (69.75±0.54) 72.50±0.53 (73.80±0.59)(74.40 ±0.46),(75.10±0.21),(75.20±0.26)℃,P < 0.05;TS (3.13±0.94),(5.19 ±0.65),(4.54±0.88),(4.67±0.60),(5.55±0.43),(5.80±0.82) MPa P < 0.05 The heat shrinking temperature after treated with photooxidation increased and higher than that in the fresh group P < 0.05 While compared with fresh group there were no marked changes in maximum stress P < 0.05 Extracted tissue protein in the photooxidation after decellular group was obviously less than that in the fresh group moreover with the time of photooxidation prolonging extract tissue protein gradually decreased. CONCLUSION Decellular treatment reduces the stability of bovine jugular tissues while tissue stability is reinforced after further treatment with photooxidation moreover it is reinforced with the prolonging of photooxidation time.
