胰岛成体干细胞的研究与应用

【摘要】 目的:根据近年来国内外有关胰岛成体干细胞的研究近况,探讨胰岛成体干细胞培养、诱导分化及应用等方面的研究进展。资料来源:应用计算机检索Medline1990-01/2005-01相关胰岛成体干细胞的文献,检索词“islet,stemcells”,并限定文献语言种类为English。同时计算机检索CBM1995-01/2005-01相关胰岛成体干细胞方面的文献,检索词为“胰岛,干细胞”,并限定语言种类为中文。资料选择:对资料进行初审,选取包括胰岛成体干细胞的文献,开始查找全文。纳入标准:①胰岛。②干细胞。排除标准:①胚胎干细胞。②综述文献、重复研究、Meta分析类文章。资料提炼:共收集到48篇关于胰岛成体干细胞的文献,纳入20篇符合标准的文献,9篇文献对胰岛成体干细胞研究方法和成果进行评价。资料综合:随着成体干细胞的研究不断深入,在许多组织内找到了具分化潜能的干细胞。干细胞的研究为多种疾病的临床治疗带来了希望。胰岛成体干细胞的分离培养、鉴定及特异的分子标志的确定和诱导分化使获得大量的胰岛细胞成为可能,进而将胰岛成体干细胞应用于临床,为完全根治糖尿病提供实验和理论基础。结论:随着胰岛成体干细胞研究的日益深入,胰岛干细胞的应用将为糖尿病的治疗带来新的曙光。

【Abstract】 OBJECTIVE: To explore the advances in culture, induction, differentiation and application of pancreatic stem cells on the basis of the latest research of pancreatic stem cells in recent years.DATA SOURCES: We searched the Medline database for literatures of pancreatic stem cells published between January 1990 and January 2005 with the "islet, stem cells" in English. Meanwhile, we also searched the CBM for related articles published from January 1995 to January 2005 with the same in Chinese. STUDY SELECTION: The literatures of pancreatic stem cells were selected primarily, and literatures on pancreatic stem cells were collected, and then full-texts were searches. Inclusion criteria were ①islet and ②stem cell. Exclusion criteria were ①embryonic stem cell, ②review, repetitive research, Meta analysis articles. DATA EXTRACTION: The literatures on pancreatic stem cells were 48. Twenty articles accorded with the inclusion criteria were included. Nine articles were on research method and result of pancreatic stem cells. DATA SYNTHESIS: With the deep research on pancreatic stem cells, stem cells with the potential of differentiation were found in tissues. The research on pancreatic stem cells brought hope for clinical therapy of many diseases. The isolation culture methods, evaluation with definition, induction and differentiation of special molecules mark made that it is feasible to get enough islet cells from pancreatic stem cells in order to apply pancreatic stem cells into clinic, and provide theoretical and experiment basis for curing diabetes completely. CONCLUSION: The application of pancreatic stem cells bring new prospect for treating diabetics with the deep research on pancreatic stem cells.
