用微卫星标记辅助鉴定几种小麦缺体—四体
Simple Sequence Repeats-Assisted Identification of Several Nulli-Tetrasomes in Wheat
【摘要】 用已准确定位到小麦染色体上的部分SSR标记辅助鉴定3个中国春缺体-四体材料:N2AT2B、N2BT2D、N2DT2A。以中国春小麦为对照,首先选用缺失染色体上的SSR引物,PCR扩增结果表明3个材料均缺失该对染色体;进一步用分布于小麦21条染色体上的SSR引物进行PCR扩增,发现除缺失染色体外的其余染色体均有扩增,说明该材料至少有其余20对染色体中的每一条;最后用缺体-四体材料对24个新开发未知位点的EST-SSRs标记进行染色体定位,结果其中10个EST-SSRs位点被分别定位在小麦2A、2B、2D染色体上。
【Abstract】 SSRs(simple sequence repeats),or microsatellites,which were already assigned to wheat chromsomes,were used to test the authenticity of 3 nulli-tetrosomes lines of wheat: N2AT2B,N2BT2D,N2DT2A.Firstly,PCR amplified products implied that they were nullisomes.Secondly,after evaluating the other chromosomes except the lost chromosomes,they were truly nulli-tetrosome lines.And the identified lines were then used to localize some newly designed and unkown EST-SSRs loci on the chromosomes.A total of 10 microsatellites loci were derived from the 10 primer pairs and separately located onto the wheat chromosomes: 2A,2B,2D.And this procedure proved to be convenient to handle for identification of Nulli-Tetrasomes in wheat.
