大骨节病儿童及成人关节软骨细胞凋亡特征的比较
The apoptotic chondrocytes in articular cartilage of children and adults with Kashin-Beck disease
【摘要】 目的探讨细胞凋亡及相关调控因子Bcl-2、Bax在大骨节病(KDB)儿童和成人关节软骨中的表达及分布的异同。方法收集15例大骨节病儿童和12例大骨节病成人,采用脱氧核糖核酸末端转移酶介导的脱氧核糖核酸缺口标记(TUNEL)技术和Bcl-2、Bax免疫组化抗生物素蛋白-生物素-碱性磷酸酶(B-SA)法染色,比较正常儿童和成人与大骨节病儿童和成人关节软骨中软骨细胞凋亡及相关调控因子Bcl-2和Bax的表达。结果KDB儿童关节软骨中层凋亡阳性细胞数较正常关节软骨显著增多(P<0.01);成人KDB关节软骨凋亡阳性细胞分布以表、中层显著(P<0.05)。②KDB儿童关节软骨表、中层Bcl-2、Bax的表达显著高于正常关节软骨中的表达(P<0.01),其中表层最高。③KDB成人关节软骨各层Bcl-2、Bax的表达显著高于正常关节软骨中的表达(P<0.05),其中表层最高。④KBD成人关节软骨各层凋亡细胞、Bcl-2、Bax阳性表达率均较KBD儿童显著增高,且Bax的表达较Bcl-2表达增多。结论KBD成人关节软骨凋亡细胞、Bcl-2、Bax阳性表达率均较KBD儿童显著增高,且Bax的表达较Bcl-2表达增多。
【Abstract】 Objective To investigate the different characteristics of chondrocyte apoptosis and the expression of(Bcl-2) and Bax in articular cartilage of children and adults with Kashin-Beck disease(KBD) so as to probe into the pathogenesis of chondronecrosis in KBD.Methods The collected samples of articular cartilage were divided into four groups: the control children(15 cases),KBD children(15 cases),the control adults(12 cases) and KBD adults(12 cases).Chondrocyte apoptosis was detected by the TUNEL method,and the Bcl-2 and Bax positive articular chondrocytes were stained by B-SA immunohistochemistry.Results ① The percentage of positive apoptotic chondrocytes stained by TUNEL in the middle zone of articular cartilage in the KBD-children group was higher than that in the control(P<0.01);the positive apoptotic chondrocytes were found throughout the articular cartilage of KBD-adults,and were prominent in the upper zone and middle zone(P<0.05).② Bcl-2 and Bax positive cells in the upper zone and middle zone of KBD-children articular cartilage were significantly higher than those in the control(P<0.01).③ Bcl-2 and Bax positive cells in the upper zone and middle zone of KBD-adults articular cartilage were significantly higher than those in the control(P<0.05).(④ Apoptotic),Bcl-2 and Bax positive cells in the upper zone,middle zone and deep zone of KBD-adults articular cartilage were significantly higher than those in KBD children,and the expression of Bax was higher than that of Bcl-2.(Conclusion)The apoptotic chondrocytes and Bcl-2,Bax positive chondrocytes in articular cartilage of adults with KBD are significantly higher than those of the children with KBD,and the expression of Bax is higher than that of Bcl-2.
