二噁致大鼠肺癌与芳香烃受体介导关系的研究

【摘要】 目的用致癌物四氧二苯二氧杂环已二烯(二,TCDD)和苯并(a)芘(B(a)P)联合诱导构建大鼠肺癌的模型,探讨芳香烃受体(AhR)介导TCDD致肺癌的发生机制。方法选Wistar大鼠80只,随机分为ABCD 4组分别代表施以TCDD、TCDD+B(a)P、B(a)P染毒的处理组和未施任何染毒处理的对照组,各组在第1.5、3、4.5和6个月,分批系列宰杀,观察肺部癌变情况。结果结果表明,TCDD染毒组在101 d时出现首例肺癌,累计TCDD用量865.94 ng,致癌率为15%,TCDD+B(a)P联合染毒组在81 d时出现首例肺癌,TCDD累计用量622.34 ng,B(a)P累计用量为26.83 mg;致癌率30%,B(a)P染毒C组在161 d时,出现首例肺癌,B(a)P累计用量为87.58 mg,致癌率为5%;对照组未出现肺癌。4组之间比较,差异有统计学意义(P<0.05)。结论TCDD和B(a)P成功地诱发了大鼠肺癌,证实了TCDD既是致癌剂又是促癌剂。

【Abstract】 Objective In order to investgate the mechanism causing lung carcinoma by 2,3,7,8-tetrahlorodibenzo-p-dioxin(TCDD),which mediaed by aryl hydrocarbon receptor(AhR),we established an animal model on rats lung carcinogenesis by carcinogen TCDD and B(a) P.Methods 80 Wistar rats were randomly divided into A,B,C experiment groups and control group D.A,B,C were treatmented with TCDD,TCDD+B(a)P,B(a)P respectively,and group D was not perfused by any carcinogen through the whole period.The rat of experiment group A,B,C and control group D were slain in the 1.5,3,4.5,6 month after exposed to TCDD and B(a)P.Results The first lung carcinoma appeared in A group when the experiment was at 101 d,the quantum of TCDD and up to 865.94 ng,and the rate of lung carcinoma in A group was 15%;the first lung carcinoma in B group was at 81 d,the quantum of TCDD add up to 622.34 ng,B(a)P add up to 26.83 mg,the rate of lung carcinoma was 30%;the first lung carcinoma in C group was at 161 d,the quantum of B(a)P add up to 87.58 mg,the rate of lung carcinoma was 5%;There was no lung carcinoma appeared in control group.the difference of lung carcinogenesis rate among 4 groups was statistic significant(P<0.05).Conclusion Model on rats lung carcinoma was successfully established by TCDD and B(a) P.This study showed conclusively that TCDD was not only a complete carcinogen,but also a cocarcinogen.
