【摘要】 目的观察转染过氧化氢酶-过氧化物酶(CPX)基因对神经毒物质β淀粉样肽(Aβ)诱发细胞凋亡的保护作用。方法转染并筛选稳定高表达CPX基因的PC12细胞系,用神经毒物质Aβ处理细胞,观察其抗逆变致凋亡的能力。通过电镜、DNA片段化的测定、MTT比色微量分析及流式细胞凋亡率分析观察转基因后对Aβ诱发细胞凋亡的保护作用。结果加入神经毒物质Aβ后,未转染CPX基因的PC12细胞出现凋亡的特征性形态学改变,电泳可见断裂的DNA片段,流式细胞仪检测细胞凋亡率较CPX基因转染组明显增加。MTT比色微量分析显示细胞活力下降(P<0.001)。结论Aβ诱导的神经细胞凋亡与活性氧的产生有关,转染CPX基因能对抗Aβ的神经毒性,保护Aβ所致的神经细胞凋亡。更多还原

【Abstract】 Objective To observe the protective effects of transfection PC₁₂ cells with catalase-peroxidase（CPX） gene against cells apoptosis induced by amyloid beta-peptide（Aβ）.Methods First transfected the PC₁₂ cells with CPX gene and selected the monocloned cell line which could express CPX gene.Then exposed the cells to Aβ,one of the neurotoxins,measured the changes of cells apoptosis by electric microscope,MTT assay,DNA electrophoretic analysis and flow cytometric DNA analysis.Results Exposure of cells to Aβ resulted in characteristic changes of apoptosis,including morphologic changes,DNA fragmentation and an increase in the apoptosis rate.MTT assay showed that cell activity was decreased remarkably（P<0.001）.While in the cells transfected with CPX genes,there were no such remarkable changes when treated with Aβ.Conclusion Aβ induced neurotoxic effect is associated with the production of ROS.Transfection of CPX gene can protect cells from apoptosis induced by Aβ.更多还原
