硒对六价铬诱导L-02肝细胞DNA损伤的影响
Effects of selenium on chromium(Ⅵ)-induced DNA damage in L-02 hepatocytes
【摘要】 目的研究硒(Se)对六价铬Cr(Ⅵ)诱导肝细胞DNA损伤的影响,探讨Se对Cr(Ⅵ)中毒预防的可能性。方法分别用8和32μmol/L重铬酸钾(K2Cr2O7)溶液处理在含有0.1和10μmol/L亚硒酸钠(Na2SeO3)中培养的L-O2细胞,采用单细胞凝胶电泳(SCGE)检测L-02肝细胞DNA损伤程度。结果8和32μmol/L K2Cr2O7单独作用时,可引起L-02肝细胞DNA损伤。当Na2SeO3与K2Cr2O7联合作用时,0.1μmol/L NaSeO3对8、32μmol/L K2Cr2O7诱导的DNA损伤存在明显的拮抗作用,而10μmol/L Na2SeO3单独存在与K2Cr2O7相互作用均具有诱导DNA损伤的作用。结论Se对L-02肝细胞DNA损伤的影响具有剂量依赖性,即Se在较低处理组水平具有降低Cr(Ⅵ)对L-02肝细胞DNA损伤的作用,在高处理组水平能诱导细胞DNA损伤。
【Abstract】 Objective To study on effects of selenium against DNA damage induced by chromium(Ⅵ)(Cr(Ⅵ)) and explore the possibility of selenium used in antagonizing to Cr(Ⅵ) induced toxicity.Methods Potassium dichromate at concentrations of 8 and 32 μmol/L was added in L-02 hepatocytes culture pretreated with sodium selenite at concentrations of 0.1 and 10 μmol/L respectively,and DNA damage in L-02 hepatocytes was detected using the single cell gel electrophoresiss(SCGE).Results Cr(Ⅵ) induced DNA damage in L-02 hepatocytes at concentrations of 8 and 32 μmol/L.Sodium selenite at concentration of 0.1 μmol/L inhibited DNA damage induced by Cr(Ⅵ),but at 10 μmol/L increased the extent of Cr(Ⅵ)-induced DNA damage.Conclusion The effect of selenium on L-02 hepatocytes DNA damage is relative to concentration of it.At lower concentration,selenium can protect L-02 hepatocytes from DNA damage induced by Cr(Ⅵ),but at higher concentration,it can induce DNA damage.
