胃电刺激对大鼠十二指肠部分肠神经递质释放的影响
Effect of Gastric Electrical Stimulation on Release of Some Neurotransmitters in Enteric Nervous System of Rat Duodenum
【摘要】 背景:胃电刺激(GES)对胃动力的影响已引起广泛关注,但关于GES后十二指肠肌间丛神经及其递质的变化,目前所知尚少。目的:研究GES对大鼠十二指肠壁内乙酰胆碱(Ach)、一氧化氮(NO)、P物质(SP)和血管活性肠肽(VIP)释放的影响。方法:建立Wistar大鼠GES模型,将模型大鼠分为GES组(n=10)和对照组(n=6)。选用适宜的刺激参数控制GES组胃慢波,应用免疫组化方法结合图像分析技术分析十二指肠肌间神经丛胆碱能、NO能、SP能和VIP能神经活性的变化。结果:GES组十二指肠肌间神经丛胆碱乙酰转移酶(ChAT)、神经元型一氧化氮合酶(nNOS)免疫反应阳性纤维较对照组明显增多、染色增强,易见ChAT、nNOS免疫反应阳性神经节和阳性神经元细胞体;而SP、VIP免疫反应阳性纤维和末梢及其染色强度在GES后无明显变化。GES组肌间神经丛ChAT、nNOS免疫反应阳性产物的平均光密度值显著高于对照组(P<0.001),SP、VIP免疫反应阳性产物的平均光密度值则与对照组无显著差异(P>0.05)。结论:GES后大鼠十二指肠壁内Ach、NO释放增多,VIP、SP则无明显变化。
【Abstract】 Background: The effects of gastric electrical stimulation (GES) on gastric motility have received much attention in recent years, but the changes of duodenal myenteric plexus and its neurotransmitters after GES remains unclear. Aims: To explore the effect of GES on release of acetylcholine (Ach), nitric oxide (NO), substance P (SP), and vasoactive intestinal peptide (VIP) in rats’ duodenum. Methods: Wistar rat models with two pairs of electrode positioned in stomach were constructed, and these rats were divided into two groups: GES group (n=10) and control group (n=6). Optimal stimulation parameters were used to entrain gastric slow wave in GES group. Changes of nerves containing choline acetyltransferase (ChAT), neuron nitric oxide synthase (nNOS), SP and VIP in duodenal myenteric plexus were studied using immunohistochemical method and quantitatively measured with computerized image analyzer. Results: The ChAT or nNOS positive nerve fibers increased significantly and their immunostaining was stronger in the duodenal myenteric plexus of the GES group than that of the control group, ChAT and nNOS positive ganglion and neuronal cell bodies could be found easily. However, the SP or VIP positive nerve fibers and endings and their immunostaining did not differ obviously between the GES and the control groups. The mean optical densities of ChAT-immunoresponse (IR) and nNOS-IR positive products in myenteric plexus were much higher in the GES group than those in the controls (P<0.001), but that of SP-IR and VIP-IR positive products had no obvious difference between GES and control groups (P>0.05). Conclusions: After GES, the release of Ach and NO is significantly increased in rat duodenum, but has no change in VIP and SP.
