羰基铼[188Re]标记含RGD多肽
Radiolabeling of RGD Peptide With 188Re-tricarbonyl Complex
【摘要】 以fac-[188Re(CO)3(H2O)3]+为前体标记了2个含RGD的多肽:HGRGD(D)F(组氨酸-甘氨酸-精氨酸-甘氨酸-天冬氨酸-苯丙氨酸)和H CRGD(D)FC(组氨酸-半胱氨酸-精氨酸-甘氨酸-天冬氨酸-苯丙氨酸-半胱氨酸)。75℃反应30 min即得到了标记产物188Re-HGRGD(D)F和H CRGD(D)FC,标记率和放化纯度均>90%。体外稳定性实验结果表明,两个标记产物在小牛血清中稳定性都差于磷酸缓冲溶液;相比之下H CRGD(D)FC稳定性稍好于188Re-HGRGD(D)F。标记产物不会引起红细胞的溶血和凝聚现象,表明这两种标记物均可用作注射用药。
【Abstract】 Two peptides containing RGD : HGRGD(D)F and H(CRGD(D)FC) are radiolabeled with fac-[188Re(CO)3(H2O)3]+.High radiolabeling ratios and chemical purity(>90%) are achieved in 30 min under the reaction condition of 75 ℃.Both of the two radiolabeled peptides are more steady in phosphate buffered saline than new-born calf serum and the stability of H(CRGD(D)FC) is better than that of 188Re-HGRGD(D)F.The hemolysis of red blood cells is not observed during incubation at 37 ℃ for 3 h.
