Cu(phen)22+与6-羟基嘌呤及DNA的相互作用
The Interaction among Cu(phen)22+,6-Mercaptopurine and DNA
【摘要】 目的为开发新的抗癌药物提供一定的理论研究依据。方法在Tris-NaCl(pH=7.2)缓冲溶液中,应用电化学实验、电子吸收光谱分析等技术研究了Cu(phen)22+(phen=1,10-邻菲咯啉)与6-羟基嘌呤(6-HX)及DNA间的相互作用。结果Cu(phen)22+与6-HX发生了较弱的相互作用,其作用产物与DNA间也存在较弱的相互作用。结论在此作用体系中Cu(phen)2+2、6-HX、Cu(phen)22+·6-HX复合物间可能存在如下平衡:Cu (phen)22++6-HX(?)Cu(phen)2(2+)·6-HX复合物,少量DNA的存在可以加速Cu(phen)22+·6-HX复合物的解离。
【Abstract】 Objective:To provide the tbeoretical basis for developing new anticancer drug.Methods:The interaction of Cu(phen)22+(phen=1,10-phenanthroline)with 6-hypoxanthine(6-HX) and DNA in Tris-NaC1 buffer solution (pH=7. 2)was investigated by electrochemistry,electron absorption spectroscopy technique.Results:Experimental results showed a weak interaction between Cu(phen)22+ complex and 6-HX,and the interaction of theit binding product with calf thymus DNA was also weak.Conclusion:The results indicate that there is an obvious interaction equilibrium among Cu(phen)22+ complex,6-HX and DNA as follows:Cu(phen)22+ + 6-HX(?)Cu(phen)22+·6-HX.Wben DNA is added to the solu- tion,it makes the interaction equilibrium move leftwards.
